慢性支气管炎动物模型制备要素和模型评价的文献分析

摘　　要：目的研究慢性支气管炎的动物模型的制备要素,为动物实验方法和模型完善提供参考依据。方法在中国知网、万方数据库、PubMed数据库中检索收集2000年1月—2022年1月慢性支气管炎动物模型的相关文献,对文献中记载的模型动物、模型种类性别、造模方式、具体造模方法等情况进行整理归纳,建立数据库进行统计分析。结果选出243篇符合纳入标准的文献,动物种属多选用SD大鼠(126篇,51.85%),其次为Wistar大鼠(70篇,28.81%),昆明小鼠(30篇,12.35%),动物性别以雄性居多(120篇,49.38%);造模方式以烟熏法(114篇,46.91%)、气管内注射脂多糖(lipopolysaccharides,LPS)及其复合因素造模方法(102篇,41.98%)为主。模型的评价标准包括模型动物一般状况、肺组织形态学观察、实验室指标和影像学检查等多个方面。结论收集分析了国内外22年来慢性支气管炎动物模型制备的文献,对文献中造模动物种属、动物性别、造模方法、造模试剂及用量、造模周期等模型制备要素进行整理归纳,同时对动物模型造模方法及成功造模后的检测指标进行评价,以期构建出重现性好、成功率高与临床疾病吻合的动物模型,为提高动物模型构建效率、科学评价动物模型提供参考。Objective To study the preparation elements of animal models of chronic bronchitis and provide a reference basis for the improvement of animal experimental methods and models.Methods Relevant literature on animal models of chronic bron-chitis from January 2000 to January 2022 was collected by searching in China Knowledge Network,Wanfang database and PubMed database.The model animals,model species gender,modelling methods and specific modelling methods recorded in the literature were collated and summarized,and a database was established for statistical analysis.Results It selected 243 articles that met the inclusion criteria,with the majority of animal species selected being SD rats(126 articles,51.85%),followed by Wistar rats(70 articles,28.81%)and Kunming mice(30 articles,12.35%).The modelling methods were mainly fumigation(114 articles,46.91%),intratracheal injection of lipopolysaccharides(LPS)and its combination of factors(102 articles,41.98%).The evalu-ation criteria of the models included the general condition of the model animals,lung histomorphological observations,laboratory parameters and imaging.Conclusion This paper collected and analyzed the literature on the preparation of animal models for chronic bronchitis over 22 years at home and abroad,and summarized the model preparation elements such as modeling animal species,animal gender,modeling method,modeling reagents and dosage,and modeling period in the literature,as well as evaluated the modeling methods and testing indexes after successful modeling,in order to construct animal models with good reproducibility,high success rate and clinical disease match,and provided reference for improving the efficiency of animal model construction and scientific evaluation of animal models.
