摘 要:目的基于“谱效”关系研究金银花药材抑菌作用及活性成分。方法采用HPLC测定商品金银花主要基原种忍冬、灰毡毛忍冬、细毡毛忍冬的化学成分,建立3种药材指纹图谱,以6种常见致病菌MRSA、MSSA、铜绿假单胞菌、鲍曼不动杆菌、大肠杆菌、肺炎克雷伯菌为指示菌,以抑菌前后的600nm下的A值计算的抑菌率为药效指标,使用灰色关联度分析法结合偏最小二乘法构建忍冬、灰毡毛忍冬、细毡毛忍冬谱效关系。结果忍冬、灰毡毛忍冬、细毡毛忍冬共检测出16个成分峰,对照品指认出9个成方峰;忍冬、灰毡毛忍冬、细毡毛忍冬对MRSA、MSSA、铜绿假单胞菌、鲍曼不动杆菌具有一定的抑制作用;16(异绿原酸C)、13(异绿原酸A)、7号峰为3者抑制MRSA的主要成分;3(绿原酸)、6(咖啡酸)、7号峰为3者抑制MSSA的主要成分;12(异绿原酸B)、7号峰为3者抑制铜绿假单胞菌的主要成分;3(绿原酸)号峰为抑制大肠杆菌的主要成分;16(异绿原酸C)号峰为抑制肺炎克雷伯氏菌的主要成分。结论通过谱效关系研究明确了忍冬、灰毡毛忍冬、细毡毛忍冬对多种致病菌具有抑制效果,其抑菌作用是多种成分共同作用的结果,为金银花不同基原种药材的药效物质基础研究与应用开发提供参考。Objective To study the spectrum-effect relationship of the antibacterial effects of Lonicerae Japonicae Flos based on grey relational degree method and partial least squares method.Methods High performance liquid chromatography was used to es-tablish the fingerprints of Lonicera japonica Thunb.Lonicera macranthoides Hand.-Mazz.and Lonicera similis Hemsl,and six common pathogenic bacteria Methicillin-resistant Staphylococcus aureus,Staphylococcus aureus,P.Aeruginosa,Acinetobacter baumannii,Escherichia coli,Klebsiella pneumoniae was used as the indicator bacteria,and the antibacterial rate was calculated by the OD600nm value before and after the antibacterial effect.effective relationship.Results A total of 16 component peaks were detected in Lonicera japonica Thunb.Lonicera macranthoides Hand.-Mazz.and Lonicera similis Hemsl,and 9 peaks were iden-tified by the reference substance.Acinetobacter baumannii has a certain inhibitory effect;peaks 16(isochlorogenic acid C),13(isochlorogenic acid A),and 7 are the main components that inhibit MRSA;3(chlorogenic acid),6(caffeic acid)and peak 7 are the main components that inhibit MSSA;peaks 12(isochlorogenic acid B)and 7 are the main components that inhibit P.Aeruginosa;peak 3(chlorogenic acid)is the main component that inhibits Escherichia coli;the peak 16(isochlorogenic acid C)is the main component that inhibits Klebsiella pneumoniae.Conclusion Through the spectrum-effect relationship study,it is clear that Lonicera japonica Thunb.Lonicera macranthoides Hand.-Mazz.and Lonicera similis Hemsl have inhibitory effects on various pathogens.Provide reference for the basic research and application development of Lonicera japonica Thunb.Lonicera macranthoides Hand.-Mazz.and Lonicera similis Hemsl.
