摘 要:为探究牛磺酸对高温条件下奶牛乳腺上皮细胞增殖和凋亡的影响,本试验在奶牛乳腺上皮细胞中添加不同浓度的牛磺酸进行42℃高温处理,通过CCK8检测细胞活力,Annexin V-FITC检测细胞凋亡水平,利用RT-PCR检测细胞中凋亡相关基因mRNA的表达变化。结果表明,高温条件下8,32mmol/L牛磺酸能促进奶牛乳腺上皮细胞增殖,缓解细胞凋亡;奶牛乳腺上皮细胞中Caspase-3、Caspase-8、Caspase-9、Bax、Bax/Bcl-2基因的表达量随牛磺酸浓度的增加先降低后升高,抗凋亡基因Bcl-2表达量变化则呈相反趋势,Caspase-3、Caspase-9、Bax基因表达量及Bax/Bcl-2在32mmol/L组最低,与对照组显著差异(P<0.05);Caspase-8基因表达量在0~128mmol/L处理组间差异不显著(P>0.05),Bcl-2基因在32mmol/L处理组的表达量显著高于其他处理组(P<0.05),Caspase-3、Caspase-8、Caspase-9、Bax/Bcl-2在256mmol/L处理组的表达量较对照组显著升高(P<0.05)。说明一定浓度的牛磺酸可以提高奶牛乳腺上皮细胞的增殖和抗凋亡能力,从而减轻了高温对细胞的损伤。This study aims to investigate the effect of taurine on the proliferation and apoptosis of bovine mammary epithelial cells under high temperature conditions.The experiment was performed by adding different concentrations of taurine to bovine mammary epithelial cells with high temperature at 42℃.The cell viability was detected by CCK8,the apoptosis level was detected by Annexin V-FITC,and mRNA expression of apoptosis-related genes in the cells were measured by RT-PCR.The results showed that 8and 32mmol/L taurine promoted bovine mammary epithelial cells proliferation and alleviated apoptosis.The expression levels of Caspase-3,Caspase-8,Caspase-9,Bax genes and Bax/Bcl-2ratio in bovine mammary epithelial cells first decreased and then increased with the increase of taurine concentration,while the expression of anti-apoptosis gene Bcl-2showed the reverse change trend.The expression levels of Caspase-3,Caspase-9,Bax genes and Bax/Bcl-2 was the lowest in 32mmol/L group,which was significantly different compared with the control group.The expression level of Caspase-8gene had no significant difference between 0-128mmol/L treatment groups(P>0.05).Compared with other group,the expression of Bcl-2gene in 32mmol/L group was significantly improved(P>0.05).The expression of Caspase-3,Caspase-8,Caspase-9and Bax/Bcl-2ratio in 256mmol/L group was significantly higher than that of the control group(P>0.05).Taurine can ameliorate the proliferation and anti-apoptotic ability of bovine mammary epithelial cells,and reduced the damage of cells under high temperature condition.
