摘  要:目的通过分析尼古丁依赖患者肾嫌色细胞癌组织中c-kit的表达情况,并对差异表达的c-kit进行相关生物信息学分析,进一步探讨其在尼古丁依赖肾嫌色细胞癌患者中的潜在作用机制。方法利用癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库,分析肿瘤组织与癌旁组织中c-kit表达差异,meta分析验证上述结果。基于UALCAN数据库,分析影响基因表达的临床病理特征。通过Linkedomics进行c-kit共表达基因的基因富集通路及PPI网络图的获取。Timer分析c-kit表达与肿瘤免疫浸润的关系。免疫组化分析尼古丁依赖肾嫌色细胞癌患者肾组织中c-kit蛋白的表达情况。结果c-kit在肾嫌色细胞癌组织中异常高表达,且表达与患者肿瘤分期相关。KIT在肾嫌色细胞癌的免疫浸润中发挥着重要的作用。吸烟能诱导肾嫌色细胞癌患者肿瘤组织中c-kit蛋白表达。结论尼古丁作为肾癌的重要诱发因素,可能通过激活肿瘤组织中c-kit活性在肾嫌色细胞癌的发生发展过程中发挥重要作用。Objective To explore the potential mechanism of c-kit in nicotine dependent chromophobe renal cell carcinoma(ChRCC)patients by analyzing the expression of c-kit in ChRCC tissues and the function of c-kit and associated genes through bioinformatics methods.Methods The expression of c-kit in cancer tissues and adjacent tissues was analyzed via using The Cancer Cenome Atlas(TCGA)database,and the above results were verified by meta-analysis.Based on the UALCAN database,the clinicopathological characteristics associated with c-kit were analyzed.The gene enrichment pathways of c-kit co-expressed genes and PPI network diagram were obtained through Linkemics.The relationship between c-kit expression and tumor immune infiltration was analyzed by Timer.The expression of c-kit protein in nicotine dependent ChRCC was analyzed by immunohistochemistry.Results The expression of c-kit was significantly high in ChRCC,and its expression was correlated with tumor staging.c-kit plays an important role in the immune infiltration of ChRCC.Smoking could induce the c-kit protein expression in ChRCC.Conclusion Nicotine,as an important inducing factor of renal cell carcinoma,may play a significant role in the occurrence and development of ChRCC by activating c-kit activity in tumor tissue.
