摘  要:目的探讨不同饮食诱导的非酒精性脂肪肝病(NAFLD)小鼠肝组织中脂滴包被蛋白(Plin)家族的表达谱及瘦素基因敲除对其的影响。方法实验选用8周龄雄性C57BL/6小鼠53只,按照随机数字表法分组:CDAA对照组和模型组(9周组和15周组)、WD对照组和模型组(2周组、6周组和30周组),除CDAA9周对照组为5只,其余每组6只。选用4周龄雄性ob/ob小鼠12只,按照随机数字表法分组:对照组和模型组,每组6只。对照组给予正常饮食喂养;分别采用胆碱缺乏、L-氨基酸替代蛋白饲料(CDAA)和西方饮食(WD)诱导建立野生型C57BL/6小鼠NAFLD模型;采用WD饮食诱导建立瘦素基因敲除小鼠(ob/ob)NAFLD模型。实时荧光定量PCR法检测肝组织中Plin 2,Plin 3,Plin 4,Plin 5 mRNA的表达,比较不同组之间的差异及瘦素敲除对其的影响。结果正常野生型C57BL/6小鼠的肝组织中可检测出Plin 2,Plin 3,Plin 4,Plin 5四种亚型mRNA的表达;其中Plin 2是主要的亚型,其次为Plin 3、Plin 5,Plin 4的表达量最低。在CDAA饮食诱导的NAFLD模型中,Plin 2表达无显著变化;Plin 3表达在造模15周降低(P<0.05);Plin 4表达在造模9周和15周均升高(P<0.05);Plin 5表达在造模9周和15周均降低(P<0.05)。在WD饮食诱导的NAFLD模型中,野生型小鼠肝组织中Plin 2表达在造模30周增加(P<0.05);Plin 3、5表达无显著变化;Plin 4表达在造模2周、6周和30周均升高(P<0.05)。与野生型小鼠不同,在WD饮食诱导的(ob/ob)小鼠NAFLD模型中,肝脏中Plin 2、Plin 3、Plin 5表达降低(P<0.05);而Plin 4表达无显著变化。结论Plins基因表达谱在不同饮食诱导的NAFLD小鼠肝组织中存在显著差异;瘦素基因敲除抑制NAFLD小鼠肝组织中Plins基因的表达。Objective To investigate the expression profile of the Perilipin(Plin)family and the effect of leptin gene knockout on Plins in the liver tissues of non-alcoholic fatty liver disease(NAFLD)models induced by different diets.Methods A total of 538-week-old male C57BL/6 mice were selected and divided into CDAA control group and model group(9-week group and 15-week group),WD control group and model group(2-week group,6-week group and 30-week group).There were 6 mice in each group except the CDAA 9-week control group(n=5).Twelve 4-week-old male ob/ob mice were divided into control group and model group according to the random number table method,with 6 mice in each group.The control group was fed normal diet,NAFLD models in wild-type mice were induced by choline-deficient,L-amino acid-defined(CDAA)diet and western diet(WD),respectively.NAFLD model in leptin gene knockout mice(ob/ob)was induced by WD.Real-time PCR was used to detect the expression of Plin 1-5 in liver tissues.The differences of Plin 2,Plin 3,Plin 4,Plin 5 expression in different groups and the effects of leptin knockout on Plin 2,Plin 3,Plin 4,Plin 5 were analyzed.Results Plin 2,Plin 3,Plin 4,Plin 5 mRNA were detected in normal liver tissue of wild type mice.Plin 2 was the most dominant subtype,followed by Plin 3 and Plin 5,and the expression of Plin 4 was the lowest.In the CDAA-induced NAFLD model,there was no significant change in Plin 2.The expression of Plin 3 decreased at 15 weeks after modeling(P<0.05).The expression of Plin 4 increased at 9 weeks and 15weeks(P<0.05).The expression of Plin 5 decreased at 9 weeks and 15 weeks(P<0.05).In the WD diet-induced NAFLD model,the expression of Plin 2 in liver tissue of wild-type mice increased at 30 weeks after modeling(P<0.05).There were no significant changes in Plin 3 and Plin 5.The expression of Plin 4 increased at 2 weeks,6weeks and 30weeks(P<0.05).Different from wild-type mice,the expressions of Plin 2,Plin 3 and Plin 5 in liver of WD diet-induced(ob/ob)mice in NAFLD model decreased(P<0.05).The expr
