摘　　要：目的:探讨肺鳞状细胞癌中解聚素-金属蛋白酶17(Human A disintegrin and metalloprotease 17,ADAM17)下调对自然杀伤细胞(Natural killer cell,NK)杀伤作用的影响。方法:体外培养人NCI-H520细胞株,分为正常对照组(未转染)、阴性对照组(转染nonsense siRNA)和ADAM17下调组(转染ADAM17 siRNA)。qRT-PCR和Western blotting检测各组细胞ADAM17 mRNA和蛋白表达。制备NK细胞,加入各组NCI-H520细胞中,Western blotting检测各组细胞γ干扰素(Interferon-γ,IFN-γ)、颗粒酶B(Granzyme B,GraB)、穿孔素(Pore-forming protein,PFP)蛋白表达。结果:与正常对照组相比,阴性对照组ADAM17 mRNA和蛋白表达无明显差异,ADAM17下调组ADAM17 mRNA和蛋白表达减少(P<0.01);与正常对照组相比,阴性对照组NCI-H520细胞生存率无明显差异,ADAM17下调组NCI-H520细胞生存率降低(P<0.01);与正常对照组相比,阴性对照组IFN-γ、GraB、PFP蛋白表达均无明显差异,ADAM17下调组IFN-γ、GraB、PFP蛋白表达上调(P<0.01)。结论:肺鳞状细胞癌NCI-H520细胞中ADAM17下调能增强NK细胞的杀伤作用。Objective:To investigate the effect of ADAM17 down-regulation on killing effect of natural killer cells(NK)in lung squamous cell carcinoma.Methods:Human NCI-H520 cell lines were cultured in vitro and they were divided into normal control group(non-transfected),negative control group(transfected nonsense siRNA)and ADAM17 down-regulated group(transfected ADAM17 siRNA).Real-time quantitative fluorescent polymerase chain reaction(qRT-PCR)was used to detect ADAM17 mRNA expression in NCI-H520 cells of each group,and Western blotting was used to detect ADAM17 protein expression in NCI-H520 cells of each group.NK cells were prepared and added into NCI-H520 cells of each group to detect the killing activity of NK cells.Western blotting detected the expression ofγinterferon(IFN-γ),granulozyme B(GraB)and perforin(PFP)proteins in the cells of each group.Results:Compared with normal control group,the ADAM17 mRNA and protein relative expression levels of negative control group were with no significant differences;and ADAM17 down-regulated group significantly decreased the mRNA and protein relative expression levels of ADAM17(P<0.01);ompared with normal control group,the survival rate of NCI-H520 had no significant differences;the survival rate of NCI-H520 in ADAM17 down-regulated group were significantlly decrased(P<0.01);compared with normal control group,the expression of IFN-γ、GraB、PF in negative control group had no significant differences,and the relative expression levels of IFN-γ,GraB and PFP protein were significantly increased(P<0.01).Conclusion:ADAM17 down-regulation in NCI-H520 lung squamous cell carcinoma can enhance the killing effect of NK cells.
