摘  要:目的:探讨紫花牡荆素对宫颈癌HeLa细胞miR-342-3p表达和糖酵解的影响,验证紫花牡荆素是否通过上调miR-342-3p表达抑制宫颈癌HeLa细胞糖酵解。方法:使用不同浓度(0,10,30,100 nmol/L)的紫花牡荆素处理HeLa细胞,采用紫花牡荆素(30 nmol/L)联合miR-342-3p模拟物或抑制物转染HeLa细胞,随后用实时荧光定量PCR分析HeLa细胞miR-342-3p的表达水平,通过测定相对葡萄糖消耗量和相对乳酸产物,分析紫花牡荆素及紫花牡荆素联合miR-342-3p模拟物或抑制物转染对宫颈癌HeLa细胞糖酵解的影响。结果:紫花牡荆素以浓度依赖方式上调HeLa细胞miR-342-3p表达水平,同时降低HeLa细胞的葡萄糖消耗量和抑制乳酸产生。此外,miR-342-3p模拟物协同紫花牡荆素抑制HeLa细胞的葡萄糖消耗和乳酸产生;相反miR-342-3p抑制物减弱紫花牡荆素上调miR-342-3p表达水平的作用和减弱紫花牡荆素抑制HeLa细胞的葡萄糖消耗和乳酸产生作用。结论:紫花牡荆素通过上调miR-342-3p表达水平抑制HeLa细胞糖酵解。Objective:To explore the effects of Casticin on the expression of miR-342-3p and glycolysis in cervical cancer HeLa cells,and to test whether Casticin inhibited glycolysis in cervical cancer HeLa cells by upregulating the expression of miR-342-3p.Methods:The method employed in the study was to treat HeLa cells with different concentrations(0,10,30,100 nmol/L)of Casticin and Casticin(30 nmol/L)combined with miR-342-3p mimics or inhibitor transfection.RT-qPCR assay was adopted to examine the expression level of miR-342-3p in HeLa cells.Simultaneously,through measuring relative glucose consumption and relative lactate production,the effects of Casticin or Casticin combined with miR-342-3p mimics or inhibitor transfection on the glycolysis in cervical cancer HeLa cells were detected and analyzed.Results:Casticin upregulated the expression level of miR-342-3p in a concentration dependent manner as well as reduced the glucose consumption and inhibited the lactate production in HeLa cells.Moerover,miR-342-3p mimic synergistically inhibits glucose consumption and lactate production in HeLa cells in combination with Casticin.On the contrary,miR-342-3p inhibitors could rescue Casticin s upregulation of miR-342-3p expression level as well as the inhibitory effects on glucose consumption and lactate production of HeLa cells.Conclusion:The conclusion indicated that Casticin inhibited glycolysis in HeLa cells through upregulating the expression level of miR-342-3p.
