摘 要:目的了解miR-216-5p在胰腺导管腺癌中的表达和意义。方法实时定量聚合酶链反应检测10例新鲜胰腺导管腺癌组织标本及对应的癌旁组织标本中miR-216和PAK1的表达。同时检测miR-216和PAK1在胰腺癌细胞系和正常胰腺导管上皮细胞中的表达。胰腺癌细胞系转染相应miRNA合成物后使用定量聚合酶链反应和蛋白免疫印迹检测PAK1表达水平的变化。细胞增殖实验检测miR-216介导的PAK1表达对吉西他滨对胰腺癌细胞增殖的影响。结果PAK1和Has-miR216-5p在胰腺癌病人术后标本中的表达:胰腺癌肿瘤组织中PAK1的相对表达量为(2.263±0.748),明显高于胰腺癌癌旁正常组织的(1.094±0.302),差异具有统计学意义(P<0.01)。且胰腺癌肿瘤组织中miR216-5p的相对表达量为(0.243±0.048),明显低于胰腺癌癌旁正常组织的(1.144±0.302),差异具有统计学意义(P<0.01)。结论miR-216-5p可显著抑制PAK1的表达,且miR216-5p可通过抑制PAK1的表达来协同吉西他滨的肿瘤抑制作用。Objective To investigate the expression and significance of miR-216-5p in pancreatic ductal adenocarcinoma.Methods The expression of PAK1 and miRNA-216 mRNA in ten cases of newly collected cancerous tissue and adjacent normal tissue were quantified using quantitative RT-PCR.Meanwhile,the expression of PAK1 and miRNA-216 were also investigated in PDA cell line and normal pancreatic duct epithelial cell.The variation of PAK1expression was determined by RT-PCR and Western Blot in pancreatic cancer cell line after been transfected relevant miRNA compound.The effect of miR-216 mediated PAK1 expression on the proliferation of pancreatic cancer cells was detected by cell proliferation assay.Results The relative expression of PAK1 and Has-miR216-5p in postoperative samples of patients with PDA:The relative expression of PAK1 in pancreatic cancer tissues was 2.263±0.748,which was significantly higher than 1.094±0.302 in pancreatic cancer adjacent normal tissues,and the difference was statistically significant(P<0.01).The relative expression of miR216-5p in pancreatic cancer tissues was 0.243±0.048,which was significantly lower than 1.144±0.302 in pancreatic cancer adjacent normal tissues,and the difference was statistically significant(P<0.01).Conclusions miR-216-5p can inhibit the expression of PAK1 in pancreatic cancer cell.Moreover,miR-216-5p enhanced the anti-cancer effect of gemcitabine via inhibit the expression of PAK1.
