摘 要:目的分析hsa_circRNA6448-14对食管鳞癌(esophageal squamous cell carcinoma,ESCC)细胞恶性增殖能力的影响,探讨其临床意义。方法建立hsa_circRNA6448-14敲低的ESCC KYSE30及KYSE150细胞系,实验分为对照组及观察组。通过qRT-PCR检测各组细胞系中hsa_circRNA6448-14的相对表达量;采用CCK8法及平板克隆实验检测各组细胞的体外增殖能力。通过t检验比较两组细胞间hsa_circRNA6448-14表达量差异及体外增殖能力差异。结果与对照组相比,观察组细胞中hsa_circRNA6448-14的相对表达量显著降低,体外增殖能力显著减弱,差异均具有统计学意义(P<0.05)。结论hsa_circRNA6448-14为ESCC细胞恶性增殖过程中的关键调控因子,靶向hsa_circRNA6448-14可能抑制ESCC恶性演进。Objective To analyze the effect of hsa_circRNA6448-14 on the malignant proliferation of esophageal squamous cell carcinoma(ESCC)cells and explore its clinical significance.Methods ESCC KYSE30 and KYSE150 cell lines with hsa_circRNA6448-14 knockdown were established and divided into control group and knockdown group.The relative expression of hsa_circRNA6448-14 in cells of each group was detected by qRT-PCR.The proliferation ability of cells in each group was detected by CCK8 assay and plate cloning assay.The differences of hsa_circRNA6448-14 expression and in vitro proliferation ability between the two groups were compared by t test.Results Compared with the control groups,the relative expression level of hsa_circRNA6448-14 in observation groups was significantly decreased(P<0.05),and the proliferation ability was significantly decreased in vitro(P<0.05),with statistical significance.Conclusion hsa_circRNA6448-14 is a key regulatory factor in the malignant proliferation of ESCC cells,and targeting hsa_circRNA6448-14 may inhibit the malignant evolution of ESCC cells.
