摘  要:目的 明确铜死亡相关长链非编码RNA(lncRNA)在三阴性乳腺癌(TNBC)中的预后价值及与肿瘤免疫微环境的关系。方法 收集肿瘤基因组图谱(TCGA)数据库中收录的TNBC患者临床信息和转录组测序数据,利用共表达分析筛选铜死亡相关lncRNA,通过Cox回归分析构建预后模型。基于所构建的预后模型,将TNBC患者分为不同风险分组,利用功能富集、免疫浸润分析,评估不同分组患者的肿瘤免疫微环境状态。结果 TCGA中筛选到TNBC铜死亡相关lncRNA 111个,Cox回归分析建立由MELTF-AS1、APTR、DHRS4-AS1、LINC02188和URB1-AS1等5个铜死亡相关lncRNA组成的TNBC预后模型。低风险组患者生存时间明显长于高风险组(P<0.05)。高风险组患者肿瘤组织中免疫抑制性细胞浸润增加。结论 基于铜死亡相关lncRNA所建立的预后模型可有效预测TNBC患者预后及评估肿瘤免疫微环境。Objective To investigate the correlation between the expression level of cuproptosis-related long non-coding RNA(lncRNA)and the prognosis of triple-negative breast cancer(TNBC),and to evaluate their relation with tumor immune microenvironment(TIME).Methods The clinical information and transcriptomic data of TNBC samples were collected from the cancer genome atlas(TCGA)database.Cuproptosis-related lncRNA were identified by co-expression analysis.Cox regression analysis was used to establish a prognostic risk model.Based on the prognostic model,patients with TNBC were divided into different risk subgroups.Subsequently,functional enrichment analysis and immune function analysis were performed to evaluate the TIME of different subgroups.Results A total of 111 cuproptosis-related lncRNA of TNBC were screened out.The risk model composed of 5 cuproptosis-related lncRNA,including MELTF-AS1,APTR,DHRS4-AS1,LINC02188 and URB1-AS1,was constructed using Cox regression analysis.TNBC patients were divided into different group according to the risk score,and the overall survival time in the low-risk group was significantly longer than that in the high-risk group(P<0.05).Furthermore,the infiltration of immunosuppressive cells in the tumor tissue was increased in the high-risk group.Conclusion A risk prediction model has been established on the basis of cuproptosis-related lncRNA in the study,which may be useful in the prediction of prognosis of TNBC patients and assessment of TIME.
