摘  要:目的 探究猪副产物中锌原卟啉(zinc protoporphyrin, ZnPP)形成差异。方法 本研究以猪副产物为研究对象,分析猪副产物间ZnPP含量的差异并以铁螯合酶(ferrochelatase,FECH)活性为指标探究产生差异的原因。采用紫外光谱、荧光光谱及化学模拟实验测定猪副产物中ZnPP的含量、FECH的活性,探究ZnPP与FECH之间的关系。结果 由75%丙酮提取的猪副产物中红色素的紫外光谱和荧光光谱与ZnPP标准品的紫外光谱和荧光光谱高度重合,证明从副产物中所提取的红色素为ZnPP。猪副产物间ZnPP的含量存在显著性差异,其中猪肝中ZnPP含量在孵育72h达到最大值,此时测得的荧光强度高达25655.44A.U。而FECH活性测定结果表明猪肝中FECH的活性远优于其他副产物。皮尔逊(Pearson)相关性分析结果表明ZnPP与FECH之间存在极显著关系。结论 本研究进一步确定了猪肝脏具有良好的ZnPP形成能力,不仅为副产物附加值利用提供了方向,同时为ZnPP的研究提供新的思路。Objective To explore the differences in the formation of zinc protoporphyrin(ZnPP)among pig by-products.Methods Pig by-products were taken as the research objects,the differences in ZnPP content between pig by-products were analyzed,and the reasons for the differences using iron chelatase(FECH)activity as an indicator were explored.The content of ZnPP and the activity of FECH in pig by-products were determined by ultraviolet spectroscopy,fluorescence spectroscopy,and chemical simulation experiments,and the relationship between ZnPP and FECH was explored.Results The ultraviolet spectra and fluorescence spectra of red pigment in pig by-products extracted with 75%acetone were highly coincident with those of the standard ZnPP,which proved that the red pigment extracted from the by-product was ZnPP.There was a significant difference in the content of ZnPP among pig by-products,with the ZnPP content in pig liver reaching its maximum value after 72 hours of incubation,and the fluorescence intensity measured at this time reaching 25655.44 A.U.The research results of FECH activity measurement indicated that the activity of FECH in pig liver was far superior to other by-products.The Pearson correlation analysis results indicated a highly significant relationship between ZnPP and FECH.Conclusion This study has further confirmed that pig liver has good ZnPP forming ability,which not only provides direction for the utilization of by-product added value,but also provides new ideas for the research of ZnPP.
