摘  要:环鸟苷酸-腺苷酸合成酶(cGAS)是一种DNA传感器,其在外源性DNA的刺激下能激活干扰素基因刺激因子(STING),诱导Ⅰ型干扰素和其他促炎性细胞因子的产生;且cGAS-STIGN信号通路的激活还能通过Ⅰ型干扰素促进瘤内CD8+T细胞募集,启动抗肿瘤免疫。目前认为STING激动剂能够作为抗肿瘤治疗的重要辅助药物,有望改善结直肠癌患者的预后,尤其是免疫治疗耐药患者。cGAS-STING信号通路可以通过启动固有免疫、肿瘤免疫以及与自噬相互作用发挥抗肿瘤效应,有望成为一个有效的潜在治疗靶点。Cyclic guanosine monophosphate adenylate synthase(cGAS)is a DNA sensor that can activate stimulator of interferon genes(STING)under the stimulation of exogenous DNA,inducing the production of typeⅠinterferon and other pro-inflammatory factors;and the activation of the cGAS-STING signaling pathway can also promote the recruitment of CD8+T cells in tumors through typeⅠinterferon,initiating anti-tumor immunity.Currently,it is believed that STING agonists can serve as important adjunctive drugs for anti-tumor therapy,with the potential to improve the prognosis of colorectal cancer patients,especially those with immunotherapy resistance.The cGAS-STING signaling pathway can exert anti-tumor effects by initiating innate immunity,tumor immunity,and interacting with autophagy,and is expected to become a potential effective therapeutic target.
