摘　　要：目的探讨子宫内膜癌(EC)组织中微小RNA-25-3p(miR-25-3p)、磷酸酶和紧张素同源物(PTEN)的表达及与病理参数的关系。方法选取2020年1月~2022年11月武汉市妇幼保健院收治的97例EC患者,实时荧光定量聚合酶链式反应检测癌组织与癌旁组织中miR-25-3p、mRNA PTEN表达。通过TargetScan数据库预测miR-25-3p与PTEN关系,采用Pearson相关性分析EC组织中miR-25-3p与PTEN mRNA表达的相关性,并分析二者与EC病理参数的关系。结果与癌旁组织比较,EC组织中miR-25-3p表达升高,PTEN mRNA表达降低(P<0.05)。TargetScan数据库预测发现,miR-25-3p与PTEN的3'非翻译区存在结合位点。Pearson相关性分析显示,EC组织中miR-25-3p与PTEN mRNA表达呈负相关(r=-0.721,P<0.001)。不同国际妇产科联盟(FIGO)分期、脉管侵犯、淋巴结转移EC组织中miR-25-3p、PTEN mRNA表达比较差异明显(P<0.05)。结论EC组织中miR-25-3p高表达,PTEN mRNA低表达,与FIGO分期、脉管侵犯和淋巴结转移有关,其水平变化可为EC的临床诊疗提供参考。ObjectiveTo investigate the expression of microRNA-25-3p (miR-25-3p) and phosphatase and tensin homolog (PTEN) in endometrial cancer (EC) and to analyze their relationship with EC pathological parameters.MethodsNinety-seven EC patients admitted to Wuhan Maternal and Child Healthcare Hospital from January 2020 to November 2022 were selected, and their cancer and paraneoplastic tissues were used for real-time fluorescence quantitative polymerase chain reaction to detect miR-25-3p and mRNA PTEN expression. The TargetScan database was used to predict the relationship between miR-25-3p and PTEN. Pearson correlation was used to analyze the correlation between miR-25-3p and PTEN mRNA expression in EC tissues and to analyze their relationship with EC pathological parameters.ResultsMiR-25-3p expression increased and PTEN mRNA expression decreased in EC tissues compared with paracancer tissues (P<0.05). Based on the TargetScan database, it was predicted that miR-25-3p has a binding site to the 3' untranslated region of PTEN. Pearson correlation analysis showed that miR-25-3p expression was negatively correlated with PTEN mRNA expression in EC tissues (r=-0.721, P<0.001). MiR-25-3p and PTEN mRNA expression was significantly different in EC tissues with different federation international of gynecology and obstetrics (FIGO) stages, vascular invasion, and lymph node metastasis (P<0.05).ConclusionHigh expression of miR-25-3p and low expression of PTEN mRNA in EC tissues are associated with FIGO stage, vascular invasion, and lymph node metastasis, and the changes in their levels may provide a reference for the clinical management of EC.
