摘 要:目的探讨不同实验动物和实验方式对肾病综合征模型的影响,以期为肾病综合征的基础研究提供参考。方法采用文献计量学方法,对中国知网(CNKI)、万方(Wanfang)、PubMed等数据库近10年来发表的关于肾病综合征动物模型的文献进行统计分析,比较在建立肾病综合征模型方面不同实验动物和实验方式的优缺点。结果肾病综合征模型实验动物多选用SD大鼠(93次,67.39%)和Wistar大鼠(32次,23.19%),其次为BALB/c小鼠等,以雄性为主(89.60%);实验造模方式以尾静脉注射阿霉素为主(129次),生化检测指标以血清白蛋白、血肌酐、胆固醇、尿素氮、甘油三酯为常用指标;阳性药以泼尼松为主(49次)。结论雄性SD大鼠和Wistar大鼠是建立肾病综合征模型较理想的实验动物,在该模型中,通过尾静脉注射阿霉素这一简便有效的实验方式,可以复制出与人类微小病变型肾病相似的临床和病理特征。生化检测指标和阳性药应根据研究目的不同,选择适当的种类和剂量。为了完善肾病综合征动物模型,还需建立稳定可靠的表观评价标准,可以通过检测和观察双结合的方法,为模型改进提供依据。Objective To explore the effects of different experimental animals and experimental methods on nephrotic syndrome models,and to provide reference for the basic research of nephrotic syndrome.Methods Using the bibliometric method,we statistically analyzed the literatures on animal models of nephrotic syndrome published in the past 10 years from CNKI,Wanfang and PubMed.The advantages and disadvantages of different experimental animals and methods in establishing nephrotic syndrome models were compared.Results SD rats(93 times,67.39%)and Wistar rats(32 times,23.19%)were selected as the experimental animals of nephrotic syndrome model,followed by BALB/c mice mainly male(89.60%).The main method of the experiment was tail vein injection of Adriamycin(ADR)(129 times),followed by tail vein injection of purinomycin and intraperitoneal injection of mercuric chloride.Serum albumin,serum creatinine,cholesterol,urea nitrogen and triglyceride are commonly used as biochemical indicators.The positive drugs were mainly prednisone(49 times).Conclusion Male SD rats and Wistar rats are ideal experimental animals for establishing nephrotic syndrome models.In this model,the simple and effective experimental method of injecting ADR into the tail vein can replicate clinical and pathological features similar to human minimal variant nephropathy.Biochemical test indicators and positive drugs should be selected according to different research purposes,and appropriate types and doses should be chosen.In order to improve the animal model of nephrotic syndrome,it is necessary to establish stable and reliable epigenetic evaluation standards.The combination of detection and observation can provide a basis for model improvement.
