摘  要:目的探索ras相关C3肉毒杆菌毒素底物3(ras-related C3 botulinum toxin substrate 3,RAC3)在脑胶质瘤组织的表达情况,及其对脑胶质瘤细胞迁移与侵袭能力的影响。方法用免疫组化实验检测商丘市第一人民医院神经外科收治的57例脑胶质瘤患者组织及其癌旁组织标本中RAC3的表达情况。将脑胶质瘤细胞U87MG分为实验组和对照组,实验组U87MG细胞转染RAC3-siRNA质粒,对照组U87MG细胞转染MOCK-siRNA质粒;荧光定量PCR检测各组细胞RAC3 mRNA含量,蛋白质免疫印迹法检测各组细胞RAC3、MMP2的表达情况;Transwell检测各组细胞的迁移与侵袭能力。结果脑胶质瘤患者组织RAC3阳性率为89.47%(51/57),癌旁组织中表达率为14.04%(8/57),RAC3在脑胶质瘤组织中的表达率显著高于癌旁组织,差异具有统计学意义(P<0.01)。转染siRNA后,实验组和对照组U87MG细胞中RAC3 mRNA表达量分别为1.23±0.20和0.43±0.12,RAC3蛋白表达量分别为1.19±0.11和0.23±0.08,MMP2蛋白表达量分别为1.19±0.11和0.23±0.08,实验组细胞MMP2表达显著下降(P<0.05)。实验组和对照组U87MG细胞transwell实验侵袭细胞数分别为(22±5)和(45±8)个,迁移细胞分别为(34±6)和(90±11)个,实验组U87MG细胞迁移(P<0.05)和侵袭能力(P<0.05)显著下降。结论RAC3在脑胶质瘤组织中高表达可能与其发展的恶性程度相关,且通过调控MMP2的表达影响脑胶质瘤细胞的迁移与侵袭能力。Objective To investigate the expression of ras-related C3 botulinum toxin substrate 3(RAC3)in glioma tissues and its effect on the migration and invasion of glioma cells.Methods The expression of RAC3 in 57 glioma patients and their adjacent tissues from the First People’s Hospital of Shangqiu was detected by immunohistochemical assay.According to the experimental requirements,brain glioma cells U87MG were divided into experimental group and control group.The experimental group U87MG cells were transfected with RAC3-siRNA plasmid,and the control group U87MG cells were transfected with MOCK-siRNA plasmid.RAC3 mRNA in each group was detected by fluorescence quantitative PCR.The expressions of RAC3 and MMP2 in each group were detected by Western blot.Transwell was used to detect the migration and invasion ability of cells in each group.Results The positive rate of RAC3 in glioma patients was 89.47%(51/57 cases),and the expression rate in paracancer tissues was 14.04%(8/57 cases).The expression rate of RAC3 in glioma tissues was significantly higher than that in paracancer tissues,with statistical significance(P<0.01).After siRNA transfection,mRNA expression of RAC3 in experimental group and control group was 1.23±0.20 and 0.43±0.12,and protein expression of RAC3 was 1.19±0.11 and 0.23±0.08,respectively.The expression of MMP2 protein was 1.19±0.11 and 0.23±0.08,respectively.The expression of MMP2 in experimental group was significantly decreased(P<0.05).Transwell assay showed that the number of invasive cells in experimental group and control group U87MG cells was(22±5)and(45±8),and the number of migratory cells was(34±6)and(90±11),respectively.In experimental group,U87MG cell migration and invasion ability decreased significantly(both P<0.05).Conclusion The high expression of RAC3 in glioma tissues may be related to the malignant degree of development,and affect the migration and invasion ability of glioma cells by regulating the expression of MMP2.
