摘 要:目的探索川芎嗪对鼻咽癌CNE-2细胞增殖和迁移的作用及分子机制。方法利用0μg/mL、100μg/mL、200μg/mL和400μg/mL川芎嗪处理CNE-2细胞后,CCK-8检测CNE-2细胞增殖能力;流式细胞术分析CNE-2细胞凋亡率;划痕实验分析CNE-2细胞迁移能力变化;Western Blot检测增殖、凋亡和迁移相关通路蛋白的表达变化。结果0μg/mL、100μg/mL、200μg/mL和400μg/mL川芎嗪均可抑制CNE-2细胞增殖(P<0.05)。川芎嗪可以显著促进细胞凋亡和抑制细胞迁移(P<0.01)。川芎嗪可以提高Bax和E-cadherin蛋白表达,以及降低Bcl-2、N-cadherin、Vimentin、Snail、p-PI3K和p-Akt蛋白表达(P<0.01)。结论川芎嗪抑制鼻咽癌CNE-2细胞增殖,提高细胞凋亡水平,其分子生物学机制可能与抑制PI3K/Akt信号通路激活相关;而抑制CNE-2细胞迁移能力,则可能与N-cadherin、Vimentin和Snail蛋白表达降低密切相关。Objective To explore the effects of tetramethylpyrazine on the proliferation and migration in nasopharyngeal carcinoma CNE-2 cells and its molecular mechanism.Methods After CNE-2 cells were treated with tetramethylpyrazine 0μg/mL,100μg/mL,200μg/mL,and 400μg/mL,CCK-8 was used to detect the proliferation ability of CNE-2 cells;Flow Cytometry was used to analyze the apoptosis rate of CNE-2 cells;Scratch Assay was used to analyze the changes in the migration ability of CNE-2 cells;Western Blot was used to detect the changes in the expression of proteins in the proliferation,apoptosis,and migration-related pathways.Results Tetramethylpyrazine inhibited the proliferation of CNE-2 cells at concentrations of 0μg/mL,100μg/mL,200μg/mL and 400μg/mL(P<0.05).Tetramethylpyrazine significantly promoted apoptosis and inhibited migration(P<0.01).Tetramethylpyrazine could increase the expression of Bax and E-cadherin protein,and decrease the expression of Bcl-2,N-cadherin,Vimentin,Snail,p-PI3K,and p-Akt protein(P<0.01).Conclusion Tetramethylpyrazine inhibits the proliferation of nasopharyngeal carcinoma CNE-2 cells and improves the levels of apoptosis,and its molecular biological mechanisms may associate to be the inhibited activation of PI3K/Akt signaling pathway,and the inhibition of migration may be close related to the decreased expression of N-cadherin,Vimentin,and Snail proteins in CNE-2 cells.
