摘 要:目的:制备Nb_(2)C/3D打印氮化硅(Si_(3)N_(4))复合组织工程支架,研究其对小鼠胚胎成骨细胞MC3T3细胞增殖活性影响。方法:采用3D打印技术制备Si_(3)N_(4)支架,由Nb_(2)AlC粉末通过HCl-LiF水热原位腐蚀、剥离处理制备得到Nb_(2)C纳米片悬浮液,将Si_(3)N_(4)支架浸泡于1.5 mg/mL Nb_(2)C纳米溶液获得Nb_(2)C生物涂层,制备Si_(3)N_(4)、Nb_(2)C/Si_(3)N_(4)支架浸提液孵育小鼠胚胎成骨细胞,CCK-8检测空白组、2%DMSO阳性对照组、Si_(3)N_(4)组、Nb_(2)C/Si_(3)N_(4)组孵育的MC3T3细胞增殖活性。结果:与空白组相比,阳性对照组细胞增殖活性显著性降低(P<0.05);与Si_(3)N_(4)组比较,Nb_(2)C/Si_(3)N_(4)组的MC3T3细胞增殖活性显著提高(P<0.05)。结论:Nb_(2)C/Si_(3)N_(4)生物涂层支架能促进MC3T3细胞增殖,是一种有应用潜力的骨组织工程材料。Objective:Preparation of Nb_(2)C/3D printed silicon nitride(Si_(3)N_(4))composite tissue engineering scaffolds and investiga⁃tion on biological characteristics in vitro of Nb_(2)C MXene nanocoating.Methods:Si_(3)N_(4)scaffold was prepared by 3D printing technolo⁃gy,and Nb_(2)C nanosheet suspension was prepared by HCl⁃LiF hydrothermal in situ corrosion and stripping treatment,Nb_(2)C biocoating was obtained by soaking the Si_(3)N_(4)scaffold in 1.5 mg/mL Nb_(2)C nanosolution;Nb_(2)C/Si_(3)N_(4)scaffold extracts were prepared and filtered,and then divided into four groups with MC3T3 cell lines,including blank control,positive control with 2%DMSO,Si_(3)N_(4)and Nb_(2)C/Si_(3)N_(4)groups,and cell proliferation activity was measured by CCK⁃8.Results:Compared to blank group,the proliferation activity of MC3T3 cells was significantly increased for positive control group(P<0.05);Compared to Si3 N4 group,the proliferation activity of MC3T3 cells was significantly increased for Nb_(2)C/Si_(3)N_(4)scaffold group(P<0.05).Conclusions:Nb_(2)C/Si_(3)N_(4)composite scaffold bio⁃lating can promote the proliferation of MC3T3 cells.It is a material with potential application for bone tissue engineering.
