摘 要:目的:研究湖南产山橿根70%乙醇提取物的乙酸乙酯部位化学成分及其体外抗炎作用。方法:利用MCI、硅胶和半制备高效液相等多种色谱技术对山橿根的化学成分进行分离纯化,通过化合物的理化性质及核磁数据鉴定结构;以脂多糖(LPS)诱导的RAW 264.7巨噬细胞作为炎症模型,探究化合物对炎症因子NO、IL-6、TNF-α释放量的影响。结果:从湖南地区山橿根70%乙醇提取物的乙酸乙酯部位中分离鉴定了6个吡喃酮类化合物,分别为:(E)-5-(hydroxymethyl)-4-methoxy-6-styryl-2H-pyran-2-one(1)、5,6-dehydrokavain(2)、4-methoxy-6-(2-hydroxy-4-phenylbutyl)-2H-pyran-2-one(3)、4-methoxy-6-(11,12-dimethoxy-trans-styryl)-2-pyrone(4)、rel-,trans-3-bis-[6-(4-methoxy-2-pyronyl)]-cis-2,trans-4-diphenyl cyclobutane(5)、aniba dimer A(6)。体外抗炎实验表明,化合物1和4能有效抑制炎症因子NO、IL-6、TNF-α的释放量。结论:其中,化合物1为新化合物,化合物2~6为首次从山橿中分离得到。化合物1和4有良好的抗炎作用。Objective:To study the chemical constituents of ethyl acetate extract from the 70%ethanol extract ofLindera reflexa roots and the anti⁃inflammatory effectsin vitro.Methods:The compounds from roots ofLindera reflexawere isolated and purified by vari⁃ous chromatographic techniques,such as MCI,silica gel and semi⁃preparative liquid phase.The structures were identified by physico⁃chemical properties and NMR data.The lipopolysaccharide(LPS)induced RAW 264.7 macrophages were used as the inflammatory model to investigate the effects of the compounds on the release of inflammatory cytokines NO,IL⁃6 and TNF⁃α.Results:Six pyrones were isolated from ethyl acetate extract of 70%ethanol extract ofLindera reflexaroots and identified as(E)⁃5⁃(hydroxymethyl)⁃4⁃me⁃thoxy⁃6⁃styryl⁃2H⁃pyran⁃2⁃one(1),5,6⁃dehydrokavain(2),4⁃methoxy⁃6⁃(2⁃hydroxy⁃4⁃phenylbutyl)⁃2H⁃pyran⁃2⁃one(3),4⁃methoxy⁃6⁃(11,12⁃dimethoxy⁃trans⁃styryl)⁃2⁃pyrone(4),rel⁃,trans⁃3⁃bis⁃[6⁃(4⁃methoxy⁃2⁃pyronyl)]⁃cis⁃2,trans⁃4⁃diphenyl cyclobutane(5),aniba dimer A(6),respectively.In vitroanti⁃inflammatory experiments showed that compounds 1,4 could effectively inhibit the release of inflammatory factors NO,IL⁃6 and TNF⁃α.Conclusion:Among them,compound 1 is a new compound,compounds 2 to 6 are isolated fromLindera reflexafor the first time.Compounds 1,4 have great anti⁃inflammatory effects.
