摘  要:目的研究1565 nm非剥脱点阵激光联合侧柏叶酊干预对斑秃小鼠干扰素-γ(IFN-γ)信号与NKG2D^(+)CD8^(+)T细胞的影响。方法将50只雄性C3H/HeJ小鼠随机分为正常组、模型组、点阵激光组、侧柏叶酊组及点阵激光联合侧柏叶酊组,每组10只。除正常组外,其余组小鼠均采用环磷酰胺诱导斑秃。在造模期间,点阵激光组给予1565 nm非剥脱点阵激光治疗;侧柏叶酊组给予5%侧柏叶酊在脱毛部位轻微按摩2~3 min;点阵激光联合侧柏叶酊组给予1565 nm非剥脱点阵激光及5%侧柏叶酊按摩治疗。观察各组小鼠皮肤状态和毛发生长情况;实验第7天取血,流式细胞术检测血液中NKG2D^(+)CD8^(+)T细胞比例,Western blot法检测毛囊组织中IFN-γ及其受体IFNGR1和IFNGR2表达情况。结果模型组小鼠斑秃部位的毛发出现断裂、变细,裸露皮肤表面出现黑点;各干预组斑秃情况均较模型组轻,其中点阵激光联合侧柏叶酊组最轻。模型组血液中NKG2D^(+)CD8^(+)T细胞比例和毛囊组织中IFN-γ、IFNGR1、IFNGR2蛋白相对表达量均明显高于正常组(P均<0.05)。点阵激光组、侧柏叶酊组仅毛囊组织中IFN-γ蛋白相对表达量均明显低于模型组(P均<0.05),点阵激光联合侧柏叶酊组血液中NKG2D^(+)CD8^(+)T细胞比例和毛囊组织中IFN-γ、IFNGR1、IFNGR2蛋白相对表达量均明显低于模型组及点阵激光组、侧柏叶酊组(P均<0.05)。结论1565 nm非剥脱点阵激光联合侧柏叶酊外用可通过抑制IFN-γ信号通路和NKG2D^(+)CD8^(+)T细胞的积累,从而有效改善斑秃。Objective It is to investigate the effects of 1565nm non-exfoliated lattice laser(1565 nm)combined with tincture of Platycladus orientalis leaves(POT)on interferon-γ(IFN-γ)signal and NKG2D^(+)CD8^(+)T cells in alopecia areata(AA)mice.Methods Fifty male C3H/HeJ mice were randomly divided into normal group,model control group,1565 nm non-stripping lattice laser treatment group(lattice laser group),POT group,and lattice laser combined with POT group(combination group),with 10 mice in each group.The mice in all groups except for the normal group were given cyclophosphamide to induce AA.During the modeling period,the lattice laser group was treated with 1565 nm non-exfoliating lattice laser,the POT group was treated with 5%POT by a light massage at the depilated area for 2~3 min,and the combination group was treated with 1565 nm non-exfoliating lattice laser and 5%POT.The skin state and hair growth of mice in each group were observed;their blood was taken on the 7th day of the experiment,and the proportion of NKG2D^(+)CD8^(+)T cells in the blood was detected by flow cytometry,and the expressions of IFN-γand its receptors IFNGR1 and IFNGR2 in hair follicle tissues were detected by Western blot.Results The hairs in the AA areas of mice in the model group appeared to be broken and thinned,with black spots appeared on the surface of the bare skin;the AA of all treatment groups was milder than that of the model group,that was the mildest in the combination group.The proportion of NKG2D^(+)CD8^(+)T cells in blood and the relative expressions of IFN-γ,IFNGR1 and IFNGR2 proteins in hair follicle tissues of the model group were significantly higher than those of the normal group(all P<0.05).The relative expression of IFN-γprotein in hair follicle tissues of the lattice laser group and POT group was significantly lower than that of the model group(P<0.05),and the proportion of NKG2D^(+)CD8^(+)T cells in blood and the relative expressions of IFN-γ,IFNGR1,and IFNGR2 proteins in hair follicle tissues of the combination group
