摘 要:探究绿原酸缓解疲劳的作用机制。设置空白组、试验组、对照组,连续灌胃28 d,进行负重游泳试验评价其抗疲劳效果,分别检测相关生化指标、氧化反应指标及核因子E2相关因子(nuclear factor erythroid 2-related factor 2,Nrf2)、血红素加氧酶-1(heme oxygenase-1,HO-1)、NADPH醌氧化还原酶-1(NADPH quinone oxidoreductase-1,NQO-1)的蛋白表达,利用分子对接分析与Nrf2的结合情况。绿原酸对延长小鼠负重游泳力竭时间,提升血清葡萄糖、肌糖原、肝糖原水平,降低乳酸、尿素氮、丙二醛含量及增强超氧化物歧化酶、过氧化物酶、谷胱甘肽过氧化物酶活力有显著作用(P<0.05)。Western blot结果显示绿原酸能够上调胞核Nrf2表达,下调胞浆Nrf2表达,增加HO-1和NQO-1表达。绿原酸与Nrf2蛋白的分子结合能为-3.34 kcal/mol,亲和力较强。绿原酸有助于缓解小鼠运动疲劳,可能与Nrf2信号途径提升抗氧化能力相关。To explore the mechanism of chlorogenic acid in relieving fatigue,the blank group,test group and control group of mice were set up,the mice were irrigated for 28 consecutive days and the anti-fatigue effect was evaluated by weight-bearing swimming test of mice.The related biochemical indicators,oxidation reaction indicators and the protein expression of nuclear factor E2 related factor 2(Nrf2),heme oxygenase-1(HO-1),NADPH quinone oxidoreductase-1(NQO-1)were detected respectively;macromolecular docking was used to analyze the binding with Nrf2.Chlorogenic acid had significant effects on prolonging the exhaustion time of weight-bearing swimming in mice,increasing the levels of serum glucose,muscle glycogen and liver glycogen,reducing the contents of lactic acid,blood urea nitrogen and malondialdehyde in mice,and enhancing the activities of superoxide dismutase,peroxidase and glutathione peroxidase in mice(P<0.05).Western blot results showed that chlorogenic acid could up-regulate the expression of Nrf2 in nucleus,down-regulate the expression of Nrf2 in cytoplasm,and increase the expression of HO-1 and NQO-1 in mice.The molecular binding energy of chlorogenic acid and Nrf2 protein was-3.34 kcal/mol,and the affinity was strong.Chlorogenic acid was helpful to alleviate exercise fatigue in mice,which may be related to the Nrf2 signal pathway enhancing antioxidant capacity.
