摘  要:目的 探讨在膀胱癌细胞系T24中α-微管蛋白1C链(TUBA1C)过表达对细胞生物学功能的影响及其调控的潜在分子机制。方法 通过生物信息学方法预测TUBA1C在膀胱癌组织中的转录水平以及与患者生存期的关系。膀胱癌细胞系T24中转染pcDNA3空载体和TUBA1C过表达载体,采用RT-qPCR、CCK-8、Transwell和Western blot法检测上调的TUBA1C水平对膀胱癌细胞系T24细胞的生物学功能及PI3K/AKT信号通路的影响。结果 GEPIA网站预测TUBA1C在膀胱癌组织中的mRNA水平高于相邻的正常膀胱组织,且TUBA1C高表达的膀胱癌患者5年生存率明显低于TUBA1C低表达者。TUBA1C过表达显著促进T24细胞的增殖、迁移和侵袭能力。另外,TUBA1C过表达能够显著激活PI3K/AKT信号通路。结论 TUBA1C在膀胱癌中具有促癌基因的作用,其可能通过激活PI3K/AKT信号通路促进膀胱癌的进展。Objective To investigate the effect of TUBA1C overexpression on the biological function of bladder cancer cell line T24 and the potential molecular mechanism of TUBA1C regulation.Methods The transcriptional level of TUBA1C in bladder cancer tissues and its relationship with patients'survival time were predicted by bioinformatics methods.Bladder cancer cell line T24 was transfected with pcDNA3 empty vector and TUBA1C overexpression vector.RT-qPCR,CCK-8,Transwell and Western blot were used to detect the effects of upregulation of TUBA1C expression on the biological function and PI3K/AKT signaling pathway of bladder cancer cell T24.Results The GEPIA website predicted that the mRNA level of TUBA1C in bladder cancer tissues was higher than that in adjacent normal bladder tissues,and the five-year survival rate of bladder cancer patients with high TUBA1C expression was significantly lower than the patients with low TUBA1C expression.Overexpression of TUBA1C significantly promoted the proliferation,migration and invasion ability of T24 cells.In addition,overexpression of TUBA1C significantly activated PI3K/AKT signaling pathway.Conclusion TUBA1C has an oncogenic role in bladder cancer,which may promote the progression of bladder cancer by activating the PI3K/AKT signaling pathway.
