摘 要:目的探讨SHCBP1在胶质瘤组织中的表达水平及临床价值。方法比较TCGA数据库中胶质瘤及正常组织中SHCBP1mRNA表达水平的差异。K-M曲线分析胶质瘤SHCBP1mRNA高表达及低表达组中位生存时间的差异,Cox回归分析胶质瘤患者预后的危险因素,分析SHCBP1与免疫细胞浸润程度的相关性。胶质瘤细胞株U87根据SHCBP1是否沉默分为阴性对照组、空白组和沉默组,比较3组细胞增殖、侵袭能力的变化。结果与正常组织相比,胶质瘤组织中SHCBPImRNA表达水平升高(P<0.05)。与胶质瘤SHCBP1mRNA低表达组相比,SHCBP1mRNA高表达组中位生存时间缩短(P<0.001)。年龄、WHO分级、IDH状态、1p/19q状态、组织学类型、SHCBP1表达均是影响胶质瘤患者预后的独立危险因素(P<0.05)。胶质瘤中SHCBP1与Th2细胞、巨噬细胞成正相关性(r值分别为0.845、0.414,P<0.001),与pDC细胞成负相关性(r=-0.363,P<0.001)。与空白组和阴性对照组相比,沉默组SHCBP1蛋白和mRNA表达水平降低(P<0.05)。结论SHCBP1在胶质瘤中呈高表达状态,SHCBP1可能在一定程度上参与了胶质瘤的恶性生物学行为。Objective e This paper aims to investigate the expression of SHCBP mRNA in glioma and its clinical signifi-cance.Methods"The expression of SHCBPI mRNA in glioma tissue and normal brain tissue in TCGA database was con-ducted for comparison.K-M curves were used to analyze the difference in median survival time between groups with high and low expression of glioma SHCBPI mRNA.Cox regression was used to analyze the prognostic risk factors of glioma,and the correlation between SHCBP1 and the degree of immune cell infiltration was analyzed.The glioma cell lines U87 were divided into negative control group,blank group and silence group according to whether SHCBP1 gene was silenced or not.The changes of cell proliferation and invasion between the three groups were compared.Results Compared with normal tissue,the expression of SHCBPI mRNA in glioma tissue was increased(P<0.05).Compared with the glioma group with low SHCBP1 mRNA expression,the median survival time was shorter in the group with high SHCBP1 mRNA expres-sion(P<0.001).Age,WHO grade,IDH status,Ip/19q status,histological type and SHCBPI expression were all independent risk factors for the prognosis of glioma patients(P<0.05).In glioma,SHCBPI was positively correlated with Th2 cells and macrophages(r=0.845,r=0.414,P<0.001),and negatively correlated with pDC cells(r=-0.363,P<0.001).Compared with the blank group and the negative control group,the protein and mRNA levels of SHCBP1 in the silent group were decreased(P<0.05).Conclusion SHCBP1 is highly expressed in glioma,and SHCBP1 may be involved in the malignant biological behavior of glioma.
