摘　　要：目的石研究真核翻译起始因子5A2(eukaryotic translation initiation factor5A2,EIF5A2)在膀胱癌患者尿液中的表达,探究其在膀胱癌诊断中的应用价值及与膀胱癌病理分级和浸润状态的关系。方法利用实时荧光定量PCR(RT-PCR)技术检测膀胱癌患者(41例)和健康体检者(33例)尿液中EIF5A2 mRNA的表达,分析其在膀胱癌患者和健康体检者尿液中表达差异情况。并对膀胱癌患者尿液EIF5A2表达量进行分析,探讨其在高级别(n=28)和低级别(n=13)组中的表达差异情况。也分析了EIF5A2表达量在浸润性膀胱癌(n=20)和非浸润性膀胱癌(n=21)中的表达。结果膀胱癌患者尿液EIF5A2mRNA表达升高,与对照组比,差异有统计学意义(P<0.05);高级别与低级别膀胱癌组之间EIF5A2mRNA表达差异无统计学意义(P>0.05);浸润性与非浸润性膀胱癌组之间的表达量差异无统计学意义(P>0.05)。结论EIF5A2mRNA在膀胱癌患者尿液中表达量升高,其在膀胱癌诊断中具有一定的价值,提示EIF5A2可能在膀胱癌的发生、发展过程中发挥一定作用。EIF5A2与膀胱癌的病理分级和浸润状态未见相关。Objective This paper aims to study the expression of eukaryotic translation initiation factor 5A2(EIF5A2)in the urine of patients with bladder cancer,so as to explore its application value in the diagnosis of bladder cancer and its relationship with the pathological grade and invasive status of bladder cancer.Methods Real-time fluorescence quantitative PCR(RT-PCR)was used to detect the expression of EIF5A2 mRNA in the urine of bladder cancer patients(41 cases)and healthy sub-jects(33 cases)and analyze the difference in expression of EIF5A2 mRNA in urine of bladder cancer patients(n=41)and healthy subjects(n=33).The expression of EIF5A2 in urine of patients with bladder cancer was analyzed to investigate the difference between the high-grade(n=28)and low-grade(n=13)groups.The expression of EIF5A2 in invasive bladder cancer(n=20)and non-invasive bladder cancer(n=21)was also analyzed.Results Urinary EIF5A2 mRNA expression in patients with bladder cancer was higher than that in control group,and the difference was statistically significant(P<0.05).There was no statistical significance on the difference in EIF5A2 mRNA expression between high-grade and low-grade bladder cancer groups(P>0.05).There was no statistical significance on the difference in the expression between invasive and non-invasive bladder cancer groups(P>0.05).Conclusion The expression of EIF5A2 mRNA is increased in urine of the patients with carcinoma of urinary bladder,which has definite reference value in the diagnosis of bladder cancer,suggesting that EIF5A2 may be involved in the occurrence and development of urinary bladder cancer.In addition,the expression of EIF5A2 is not related to the pathological grade and invasive status of bladder cancer.
