摘 要:目的 探讨长链非编码RNA(lncRNA)DNAJC3-AS1、微RNA-214-3p(miR-214-3p)在结肠癌组织、结肠腺瘤性息肉中的表达水平及临床意义。方法 lnCAR数据库检索结肠癌组织、正常结肠组织中LncRNA DNAJC3-AS1表达情况;选取2016年5月至2019年1月北京市丰台中西医结合医院收治的86例结肠癌病人、53例结肠腺瘤性息肉病人为研究对象,收集结肠癌病人的结肠癌组织、对应癌旁组织及结肠腺瘤性息肉病人的结肠腺瘤性息肉,实时荧光定量PCR(qRT-PCR)法测定组织中lncRNA DNAJC3-AS1、miR-214-3p表达水平;分析结肠癌组织lncRNA DNAJC3-AS1、miR-214-3p表达水平与结肠癌病人临床病理特征、预后关系;分析结肠癌组织中lncRNA DNAJC3-AS1表达水平与miR-214-3p相关性,生物信息学预测lncRNA DNAJC3-AS1与miR-214-3p的关系。结果 lnCAR数据库分析显示,结肠癌组织lncRNA DNAJC3-AS1表达水平(4.40±0.49)高于正常结肠组织(4.00±0.36)(P<0.05)。qRT-PCR检测显示,与结肠腺瘤性息肉(1.71±0.57、0.67±0.22)相比,结肠癌组织lncRNA DNAJC3-AS1表达水平(2.63±0.88)升高(P<0.05),miR-214-3p表达水平(0.31±0.10)降低(P<0.05);与癌旁组织(1.04±0.35、1.01±0.34)相比,结肠癌组织lncRNA DNAJC3-AS1表达水平升高(P<0.05),miR-214-3p表达水平降低(P<0.05)。结肠癌组织lncRNA DNAJC3-AS1、miR-214-3p表达水平在不同分化程度、淋巴结转移、血管浸润、TNM分期方面,分布差异有统计学意义(P<0.05);结肠癌组织中lncRNA DNAJC3-AS1表达水平与miR-214-3p呈负相关(r=-0.58,P<0.05),且lncRNA DNAJC3-AS1与miR-214-3p存在结合位点;lncRNA DNAJC3-AS1高表达组29.75个月、miR-214-3p低表达组30.16个月结肠癌病人累积生存率低于lncRNA DNAJC3-AS1低表达组34.69个月、miR-214-3p高表达组34.37个月(P<0.05)。结论 癌旁组织、结肠腺瘤性息肉、结肠癌组织中lncRNA DNAJC3-AS1表达依次升高,miR-214-3p表达依次降低,且结肠癌组织中lncRNA DNAJC3-AS1表达水平与miR-214-3p、临床�Objective To investigate the expression levels and clinical significance of long non-coding RNA(lncRNA)DNAJC3-AS1 and microRNA-214-3p(miR-214-3p)in colon cancer(CC)tissue and colon adenomatous polyps.Methods The lnCAR database was retrieved to search the expression of LncRNA DNAJC3-AS1 in CC tissue and normal colon tissue.A total of 86 patients with CC and 53 patients with colonic adenomatous polyps admitted to Fengtai Hospital of Integrated Traditional Chinese and Western Medicine from May 2016 to January 2019 were taken as the research objects.The CC tissue of CC patients,the corresponding paracancerous tissue and colonic adenomatous polyps of patients with colonic adenomatous polyps were collected.The expression levels of lncRNA DNAJC3-AS1 and miR-214-3p in tissues were determined by real-time quantitative PCR(qRT-PCR)method.The relationship between the ex⁃pression levels of lncRNA DNAJC3-AS1 and miR-214-3p in CC tissues and the clinicopathological characteristics and prognosis of CC patients was analyzed.The correlation between the expression level of lncRNA DNAJC3-AS1 and miR-214-3p in CC tissues was ana⁃lyzed,and the relationship between lncRNA DNAJC3-AS1 and miR-214-3p was predicted by bioinformatics.Results lnCAR database analysis showed that lncRNA DNAJC3-AS1 expression level in CC tissues(4.40±0.49)was higher than that in normal colon tissues(4.00±0.36)(P<0.05).qRT-PCR showed that lncRNA DNAJC3-AS1 expression level in CC tissues was higher(2.63±0.88)than that in colorectal adenomatous polyps(1.71±0.57,0.67±0.22)(P<0.05).The expression level of miR-214-3p was decreased(0.31±0.10)(P<0.05).Compared with para-cancerous tissues(1.04±0.35,1.01±0.34),the expression level of lncRNA DNAJC3-AS1 in CC tissues was increased(P<0.05),and the expression level of miR-214-3p was decreased(P<0.05).The expression levels of lncRNA DNAJC3-AS1 and miR-214-3p in CC tissues were significantly different in different degrees of differentiation,lymph node metastasis,vascular infil⁃tration and TNM stage(P<0.05).The e
