摘 要:目的利用生物信息学,探讨谷氨酰胺酶(glutaminase,GLS)在乳腺癌中的表达及免疫逃逸中的作用。方法运用TCGA、TIMER2.0数据库,生信预测GLS在乳腺浸润性癌中的表达情况;利用STRING数据库、LinkedOmics数据库预测在乳腺癌中GLS蛋白表达及蛋白互作作用;利用TIMER2.0数据库,分析GLS对乳腺癌免疫浸润的影响,探讨GLS对免疫检查点分子表达的影响。结果GLS mRNA在乳腺浸润性癌中表达下调,与谷氨酰胺代谢相关蛋白表达相关;GLS与免疫细胞浸润相关,且与PD1、PD-L1、CTLA-4等免疫检查点分子表达相关。结论GLS在乳腺浸润性癌中表达下调,且与肿瘤免疫细胞浸润及免疫检查点分子表达相关。Objective To explore the expression of glutaminase(GLS)in breast cancer and its role in immune escape by bioinformatics.Methods TCGA and TIMER2.0 databases were used to predict the expression of GLS in invasive breast cancer;The STRING database and LinkedOmics database were used to predict GLS protein expression and protein interactions in breast cancer;The TIMER2.0 database was used to analyze the effect of GLS on the immune invasion of breast cancer and explore the effect of GLS on the molecular expression of immune checkpoint.Results GLS mRNA was downregulated in invasive breast cancer and correlated with the expression of glutamine metabolism related proteins;GLS was associated with immune cell infiltration and expression of immune checkpoint molecules such as PD1,PD-L1,and CTLA-4,etc.Conclusion The expression of GLS was downregulated in invasive breast cancer and correlated with the infiltration of tumor immune cells and the expression of immune checkpoint molecules.
