摘  要:利用刚果红测试、分子量测定、X射线衍射以及傅里叶变换红外光谱分析黄孢原毛平革菌胞外多糖的结构,使用荧光显微镜、扫描电子显微镜和透射电子显微镜研究多糖的自组装行为。在刚果红测试中,该刚果红-多糖混合溶液最大吸收波长在低浓度碱溶液中出现明显的红移现象,并随着碱溶液浓度的提高红移现象消失,说明黄孢原毛平革菌胞外多糖具有三螺旋结构且在高浓度碱溶液中被破坏为无序结构。通过分子量测定发现,二甲基亚砜溶液中的多糖分子量明显低于水溶液中的,表明在二甲基亚砜溶液中多糖链构象发生解聚。傅里叶变换红外光谱分析发现该多糖具有多糖化合物的特征吸收峰,且含β型糖苷键。X射线衍射分析结果表明它是一种非晶体结构的多糖。荧光显微镜、扫描电子显微镜和透射电子显微镜综合分析发现多糖可形成纤维网状空间结构,并在其自组装行为下形成聚集体。以上结果表明,黄孢原毛平革菌胞外多糖是一种具有三螺旋结构的非晶体结构多糖并能自组装形成纤维状聚集体。The purpose of this study is to analyze the structure and self-assembly behavior of extracellular poly-saccharides produced by Phanerochaete chrysosporium BKMF-1767.The structure of extracellular polysaccha-rides from Phanerochaete chrysosporium is analyzed with Congo red test,molecular weight determination,X-ray diffraction and Fourier transform infrared spectroscopy,and the self-assembly behavior of the polysaccharides is studied with fluorescence microscopy,scanning electron microscopy and transmission electron microscopy.In Congo red test,the maximum absorption wavelength of the polysaccharide-Congo red solution shows obvious red-shift phenomenon in low-concentration alkali solution,and the red-shift phenomenon has disappeared with the increase of alkali solution concentration,indicating that the extracellular polysaccharides from Phanerochaete chrysosporium has a triple helix structure and the structure is destroyed into disordered structure in high concen-tration alkali solution.During molecular weight determination,the molecular weight of the polysaccharide in dimethyl sulfoxide solution is significantly lower than that in aqueous solution,indicating that the configuration of the polysaccharide chain is depolymerized in dimethyl sulfoxide solution.Fourier transform infrared spectros-copy analysis finds that the polysaccharide has characteristic absorption peaks of polysaccharide compounds and containedβ-glycosidic bonds.X-ray diffraction analysis shows that it is a kind of amorphous polysaccharide.The comprehensive analysis of fluorescence microscopy,scanning electron microscopy and transmission electron microscopy indicates that the polysaccharide can form a fibrous network-like spatial structure and form aggre-gates under its self-assembly behavior.The above results indicate that the extracellular polysaccharides from Phanerochaete chrysosporium is one kind of amorphous polysaccharide with triple helical structure,which can self-assemble into fibrous aggregates.
