摘 要:目的:研究痰湿体质人群外周血淋巴细胞亚群分布和T细胞受体(TCR)免疫组库特征谱,以揭示痰湿体质机体免疫学特征。方法:流式细胞术检测平和体质和痰湿体质志愿者外周血淋巴细胞亚群分布,利用高通量测序技术对CD8^(+)T细胞受体β链(CD8^(+)TCRβ)基因测序,进行免疫组库分析。结果:与平和体质组比较,痰湿体质人群外周血中B淋巴细胞比率下降(P<0.05),而CD8^(+)T细胞和NKT细胞比率明显升高(P<0.05);痰湿体质组CD8^(+)TCRβ链存在2种V基因(TRBV7-4、TRBV10-2)、4种V-J组合基因(TRBV7-4_TRBJ2-7、TRBV7-2_TRBJ2-1、TRBV5-8_TRBJ2-1、TRBV7-8_TRBJ2-6)、5种V-D-J组合基因(TRBV6-5_TRBJ2-1_TRBD2、TRBV7-2_TRBJ2-5_TRBD2、TRBV4-1_TRBJ1-5_TRBD2、TRBV7-4_TRBJ2-5_TRBD2、TRBV6-8_TRBJ2-1_TRBD2)表达显著上调,2种V-J组合基因(TRBV27_TRBJ2-4和TRBV30_TRBJ2-2)及1种V-D-J组合基因(TRBV25-1_TRBJ2-5_TRBD1)表达显著下调。结论:痰湿体质外周血中淋巴细胞亚群分布和CD8^(+)T细胞受体表达谱具有体质特异性,本研究为进一步揭示痰湿体质人群免疫学特征提供了免疫组库方面的科学依据,同时为痰湿体质分子诊断及该体质易患疾病的发病机制研究提供新的研究思路。Objective:To study the distribution of peripheral lymphocyte subsets and the characteristic spectrum of T-cell receptor(TCR)immune repertoire in population with Tanshi(痰湿)constitution to uncover the immunological features of Tanshi constitution.Methods:Flow cytometry was used to analyze the distribution of peripheral lymphocyte subsets in volunteers with both the Pinghe(平和)constitution and the Tanshi constitution.High-throughput sequencing technology was employed to sequence the CD8^(+)TCRβchain(CD8^(+)TCRβ)gene,and immune repertoire analysis was conducted.Results:Compared with the Pinghe constitution group,the population with Tanshi constitution exhibited a decreased ratio of B lymphocytes in peripheral blood(P<0.05),while the ratios of CD8^(+)T cells and NKT cells were significantly elevated(P<0.05).In the Tanshi constitution group,the expression of CD8^(+)TCRβchains showed significant upregulation for 2 V genes(TRBV7-4,TRBV10-2),4 V-J combinations(TRBV7-4_TRBJ2-7,TRBV7-2_TRBJ2-1,TRBV5-8_TRBJ2-1,TRBV7-8_TRBJ2-6),and 5 V-D-J combinations(TRBV6-5_TRBJ2-1_TRBD2,TRBV7-2_TRBJ2-5_TRBD2,TRBV4-1_TRBJ1-5_TRBD2,TRBV7-4_TRBJ2-5_TRBD2,TRBV6-8_TRBJ2-1_TRBD2).Additionally,the expression of 2 V-J combinations(TRBV27_TRBJ2-4 and TRBV30_TRBJ2-2)and 1 V-D-J combination(TRBV25-1_TRBJ2-5_TRBD1)was significantly downregulated.Conclusion:The distribution of peripheral lymphocyte subsets and the expression profile of CD8^(+)TCR in the population with Tanshi constitution display constitution-specific characteristics.This study provides immunological repertoire-based scientific evidence for further understanding the immunological features of the population with Tanshi constitution.Moreover,it offers new research directions for the molecular diagnosis of Tanshi constitution and the investigation of its susceptibility mechanisms to diseases.
