摘 要:目的本研究从自噬活性变化水平的角度,来分析探讨电针缓解失神经肌萎缩的可能机制。方法应用坐骨神经离断的方法,建立了失神经肌萎缩大鼠模型。通过测定肌肉湿重比来判定电针对肌肉萎缩的延缓作用。采用组织学和免疫印迹对比观察肌纤维形态学变化、评估细胞自噬相关蛋白和肌萎缩相关蛋白的变化。结果电针干预提高了失神经肌萎缩大鼠骨骼肌湿重比值。此外,电针使失神经支配大鼠腓肠肌组织中Bcl2表达增加(P<0.05),而Bax、LC3Ⅱ/LC3Ⅰ和Atrogin-1的表达下降(P<0.05)。结论电针可能通过下调自噬水平来减轻去神经支配诱导的细胞过度死亡,以减轻骨骼肌的萎缩过程。Objective This study analyzed the possible mechanism of electroacupuncture in alleviating denervation muscular atrophy from the perspective of the change level of autophagy activity.Methods The rat model of denervation muscular atrophy was established by sciatic nerve dissociation.The wet-weight ratio of muscle was measured to determine the retarding effect of electroacupuncture on muscle atrophy.The morphological changes of muscle fibers and the changes of autophagy related proteins and muscular atrophy related proteins were evaluated by histology and Western blot.Results Electroacupuncture improved the wet weight ratio of skeletal muscle in rats with denervation muscular atrophy.In addition,electroacupuncture increased the expression of Bcl-2 in gastrocnemius of denervated rats(P<0.05),while decreased the expression of Bax(P<0.05),LC3Ⅱ/LC3Ⅰ(P<0.05)and Atrogin-1(P<0.05).Conclusion Electroacupuncture may reduce the excessive cell death induced by denervation and reduce the atrophy process of skeletal muscle by down-regulating the autophagy level.
