miR-21、miR-130a在结直肠癌组织中的表达及其临床意义

摘　　要：目的:探讨结直肠癌组织中miR-21、miR-130a的表达情况,并分析其与患者预后的关系。方法:选择2017年6月至2020年5月我院收治的80例结直肠癌患者的癌组织及癌旁组织,采用定量PCR法检测其中miR-21、miR-130a的表达水平,并分析癌组织中miR-21、miR-130a表达与患者临床病理指标及预后的关系。结果:结直肠癌组织中miR-21、miR-130a表达水平明显高于癌旁组织,P<0.05。肿瘤直径≥5 cm、Ⅲ~Ⅳ期、淋巴结转移的结直肠癌患者癌组织miR-21表达分别高于肿瘤直径<5 cm、Ⅰ~Ⅱ期及淋巴结未转移组,P<0.05。Ⅲ~Ⅳ期、淋巴结转移的结直肠癌患者癌组织miR-130a表达分别高于Ⅰ~Ⅱ期和淋巴结未转移组,P<0.05。Kaplan-Meier生存分析显示,miR-21低表达组患者总生存时间(OS)、无病生存时间(DFS)明显长于miR-21高表达组,P<0.05;miR-130a低表达组的OS、DPS明显长于miR-130a高表达组,P<0.05。结论:结直肠癌患者癌组织中miR-21、miR-130a高表达,其表达与临床分期、淋巴结情况均有关,miR-21、miR-130a高表达有望成为预后生存不佳的预测指标。Objective To investigate the expression of miR-21 and miR-13Oa in colorectal cancer tissue and analyze their relationship with the prognosis of patients.Methods Cancer tissue and adjacent tissue of 80 patients with colorectal cancer admitted to our hospital from June 2017 to May 2020 were selected.The expression levels of miR-21 and miR-130a were detected by quantitative PCR,and the relationship among the expression levels of miR-21 and miR-130a in cancer tissue,clinicopathological indicators and prognosis of patients was analyzed.Results The expression levels of miR-21 and miR-130a in colorectal cancer tissue were significantly higher than those in adjacent tissue(P<0.05).The expression levels of miR-21 in cancer tissue of colorectal cancer patients with tumor diameter≥5 cm,II~IV stage and lymph node metastasis were higher than those of patients with tumor diameter<5 cm,I~II stage and non lymph node metastasis group,respectively(P<0.05).The expression levels of miR-130a in cancer tissue of colorectal cancer patients with stage Il~IV and lymph node metastasis were higher than those in stage I~II and non lymph node metastasis group,respectively(P<0.05).Kaplan-Meier survival analysis showed that the overall survival time(OS)and disease-free survival time(DFS)of patients with low ex-pression of miR-21 were significantly longer than those with high expression of miR-21(P<0.05).The OS and DPS of low expression group of miR-130a were significantly longer than those of high expression group of miR-130a(P<0.05).Conclusion The high expression levels of miR-21 and miR-130a in cancer tissue of colorectal cancer patients are related to the clinical stage and lymph node status,and the high expression levels of miR-21 and miR-130a are expected to be the predictors of poor prognosis.
