摘  要:肌酸是合成磷酸肌酸的重要原料,也是骨骼肌能量代谢的后备来源,补充肌酸在体育运动中受到了人们的广泛关注。近年来肌酸粉的销售量呈明显上升趋势,因此寻找简便、快速、准确的肌酸检测方法是食品研发人员的主要目标。采用紫外可见分光光度法分析研究了检测健身肌酸粉中肌酸含量的影响因素,包括:体系的pH显色时间和肌酸的溶剂。在最优试验条件下,利用标准品肌酸绘制出标准曲线,可知肌酸的质量浓度在200~1000μg/mL的范围内与其吸光度线性关系良好,相关系数r=0.99537,检出限=65.60μg/mL。利用紫外可见分光光度法对市售健身肌酸粉,以体积比V(甲醇)∶V(水)=20∶80的溶液为溶剂,室温放置25 min,测定肌酸的吸光度,并计算出市售健身肌酸粉Ⅱ中肌酸的质量分数为70.00%,加标回收率为100.15%。试验结果表明,紫外可见分光光度法能够简单、快速地检测出健身肌酸粉中的肌酸。Creatine is an important material for synthesizing phosphocreatine and a backup source of energy metabolism in skeletal muscles.Supplementing creatine in sports has received widespread attention.In recent years,the sales volume of creatine powder has shown a significant upward trend,so finding a simple,fast,and accurate creatine detection method is the main goal of food research and development personnel.The factors affecting the detection of creatine content in fitness creatine powder are analyzed and studied by UV visible spectrophotometry,including the pH colorimetric time of the system and the solvent of creatine.Under the optimal experimental conditions,a standard curve is drawn by standard creatine,indicating that the mass concentration of muscle is between 200 to 1000μg/mL,and it has a good linear relationship with the volume ratio that methanol to water is 20:80,with a correlation coefficient r is 0.99537 and a detection limit is 65.60μg/mL.Using UV visible spectrophotometry to determine the absorbance of creatine in commercially available fitness creatine powder,which takes a volume ratio that methanol to water is 20:80 as the solvent and is placed at room temperature for 25 minutes,the mass fraction of creatine in commercially available fitness creatine powder II is calculated to be 70%,with a recovery rate of 100.15%.The UV visible spectrophotometry can easily and quickly detect creatine in fitness creatine powder.
