摘 要:探究硝酸银(Silver nitrate,AgNO_(3))对白鲜生长及药用成分含量的影响,为药用植物白鲜的次生代谢调控提供理论依据。该研究以白鲜种子为试验材料,待种子萌发后移至添加不同浓度AgNO_(3)的MS培养基中,处理30 d后记录白鲜幼苗的根长、侧根数、鲜重和干重净增量,以不加AgNO_(3)的MS培养基为对照(control treatment,CK),并采用高效液相色谱仪(High performance liquid chromatography,HPLC)测定白鲜幼苗中白鲜碱、梣酮、黄柏酮含量。结果表明:经10μmol/L AgNO_(3)处理后显著促进了白鲜幼苗根的生长,0.5μmol/LAgNO_(3)处理后显著提高了白鲜幼苗的侧根数、鲜重、干重净增量和黄柏酮含量,50μmol/L AgNO_(3)处理后显著提高了白鲜碱含量,1μmol/L AgNO_(3)处理后显著提高了梣酮含量。通过主成分分析,该试验筛选出的AgNO_(3)诱导子最佳浓度为0.5μmol/L。In this study,silver nitrate(AgNO_(3))was used as an elicitor to preliminarily investigate the effect on growth and medicinal components of in vitro cultured Dictamnus seedlings to provide a theoretical basis for the regulation of secondary metabolism in medicinal plants.The sterilized Dictamnus seeds were inoculated in MS medium supplemented with different concentrations of AgNO_(3).After 30 days of culture,root length and lateral root number,shoot fresh and dry weight were recorded,and the contents of dictamnine,fraxinellone and obacunone were determined by high performance liquid chromatography(HPLC).The result showed that root length was obviously increased after treatment with 10μmol/L AgNO_(3),but the number of lateral roots and shoot fresh and dry weight were promoted by 0.5μmol/L AgNO.The highest contents of medicinal components were found at different concentrations of AgNO_(3),namely,obacunone content at 0.5μmol/L AgNO_(3),dictamnine content at 50μmol/L AgNO_(3),and fraxinellone content at 1μmol/L AgNO_(3).Principal component analysis confirmed 50μmol/L as the optimal AgNO_(3) concentration.
