摘 要:为探究栝楼籽多糖的提取工艺、初级结构及降血糖活性,采用热水浸提法对栝楼籽多糖进行提取,经葡聚糖凝胶G-150纯化得到均质多糖(WHP-Ⅰ)后进行了初步表征。通过体外和体内联合法测试了栝楼籽多糖的降血糖活性。得到栝楼籽多糖的最优提取条件为提取温度80℃、提取时间100 min、料液比1∶25 (g∶m L)、浸提1次,在上述条件下,WHP-Ⅰ平均提取率为19.01%。WHP-Ⅰ的重均相对分子质量为4.765×10~4,并含有5种糖残基,WHP-Ⅰ表面较为平整,有孔状结构,且附着有细小的颗粒。WHP-Ⅰ对α-葡萄糖苷酶的半抑制浓度(IC_(50))为15.837 g/L,WHP-Ⅰ具有减缓Ⅱ型糖尿病(T2DM)引起脏器损伤的潜力,给药4周后,中剂量组M-WHP-Ⅰ[200 mg/(kg·bw)]和高剂量组H-WHP-Ⅰ[300 mg/(kg·bw)]小鼠的血糖值分别降低了32.13%和42.27%,且WHP-Ⅰ对糖尿病小鼠的肾脏和肝脏具有修复作用。In order to optimize its extraction process and investigate its primary structure and hypoglycemic activity,the homogenous polysaccharide (WHP-Ⅰ) was obtained from Trichosanthes Kirilowii Maxim.seed by hot water extraction,purified by glucan gel G-150,preliminarily characterized and analyzed for its hypoglycemic performance both in vitro and in vivo.Under the optimal extraction conditions of temperature80℃,extraction time 100 min,solid-liquid ratio 1∶25 (g∶m L),and one time extraction,the average extraction rate of polysaccharide was 19.01%.Results from characterization indicated WHP-Ⅰhad a mass-average relative molecular mass of 4.765×10~4,contained five sugar residues,and displayed a flat surface as well as a porous structure with fine particles attached.Hypoglycemic activity analysis showed that WHP-Ⅰ,with a half-inhibitory concentration (IC_(50)) against α-glucosidase of 15.837 g/L,reduced the blood glucose level by 32.13%with a dose of 200 mg/(kg·bw) and by 42.27%with a dose of 300 mg/(kg·bw)after 4 weeks of administration,indicating its potential to slow the organ damage caused by diabetes mellitus typeⅡ(T2DM).Moreover,WHP-Ⅰalso demonstrated capacity of repairing the kidney and liver of diabetic mice.
