摘 要:为了合理利用羽毛,减轻对环境的污染,本研究以鸡毛为原料,以鸡毛降解率为指标,确定合适加酶量为5000 U/g,通过单因素试验和响应面试验对角蛋白酶降解鸡毛工艺条件进行优化。试验结果表明,当加酶量为5000 U/g、酶解温度为64℃、料液比为1∶48(g/mL)、酶解时间为5 h、酶解初始pH值为8.70时,鸡毛降解率达55.10%。本研究确定了角蛋白酶酶解鸡毛最佳工艺条件,可为羽毛的开发利用提供理论依据。In order to make reasonable use of feathers and reduce environmental pollution,this study used chicken feathers as raw materials and the degradation rate of chicken feathers as an indicator.The appropriate enzyme dosage was determined to be 5000 U/g.The process conditions for keratinase hydrolysis of chicken feathers were optimized through single factor experiments and response surface method experiments.The experimental results showed that when the enzyme dosage was 5000 U/g,the enzymatic hydrolysis temperature was 64℃,the material liquid ratio was 1∶48(g/mL),the enzymatic hydrolysis time was 5 hours,and the initial pH value of the enzymatic hydrolysis was 8.70,the degradation rate of chicken feathers reached 55.10%.This study determined the optimal process conditions for the hydrolysis of chicken feathers with keratinase,which could provide a theoretical basis for the development and utilization of feathers.
