摘 要:目的:探讨当归芍药散是否可以通过AMPK/mTOR信号通路激活自噬改善高糖诱导的足细胞损伤。方法:分别将足细胞分为正常对照组、模型组、当归芍药散含药血清组、自噬抑制剂组、当归芍药散含药血清加自噬抑制剂组、自噬激动剂、AMPK抑制剂、当归芍药散含药血清加AMPK抑制剂组和AMPK激动剂,并观察各组的自噬相关蛋白、细胞活性和细胞凋亡率。结果:与对照组相比,高糖组足细胞活力和Beclin-1/LC3/p-AMPK蛋白表达均降低,细胞凋亡率和p-mTOR蛋白表达均增加(P<0.01);和高糖组相比,当归芍药散含药血清组、自噬激动组、AMPK激动剂组的细胞活力和Beclin-1/LC3Ⅱ/p-AMPK蛋白表达均提高,细胞凋亡率和p-mTOR蛋白表达均降低(P<0.01);与高糖组相比,自噬抑制剂组、AMPK抑制剂组足细胞的细胞活力和Beclin-1/LC3Ⅱ/p-AMPK蛋白表达均降低,细胞凋亡率和p-mTOR蛋白表达均增加(P<0.01)。结论:当归芍药散可以通过AMPK/mTOR通路激活自噬改善高糖诱导的足细胞损伤。OBJECTIVE To investigate whether Danggui Shaoyao Powder can activate autophagy through the AMPK/mTOR signaling pathway to improve high glucose induced podocyte injury.METHODS Podocytes were divided into several groups:normal control group,model group,Danggui Shaoyao San medicated serum group,autophagy inhibitor group,Danggui Shaoyao San medicated serum plus autophagy inhibitor group,autophagy agonist group,AMPK inhibitor group,Danggui Shaoyao San medicated serum plus AMPK inhibitor group,and AMPK agonist group.The researchers observed the levels of autophagy-related proteins,cell activity,and apoptosis rate in each group.RESULTS Compared with the control group,the cell viability and the expression of Beclin-1/LC3Ⅱ/p-AMPK protein in high glucose group decreased,while the apoptosis rate and the expression of p-mTOR protein increased(P<0.01).Compared with HG group,the cell viability and the expression of Beclin-1/LC3Ⅱ/p-AMPK protein were increased,the cell apoptosis rate and the expression of p-mTOR protein were decreased in DSS-containing serum group,autophagy stimulating group and AMPK stimulating group(P<0.01).Compared with HG group,autophagy inhibitor group and AMPK inhibitor group decreased podocyte viability and Beclin-1/LC3Ⅱ/p-AMPK protein expression,and increased apoptosis rate and p-mTOR protein expression(P<0.01).CONCLUSION Danggui Shaoyao Powder can improve podocyte injury induced by high glucose by activating autophagy through AMPK/mTOR pathway.
