摘  要:目的研究脂质激酶AGK对人胃癌细胞SGC-7901迁移和侵袭能力的影响及探索潜在的分子机制。方法培养胃癌细胞SGC-7901,用HiGene试剂转染AGK质粒,验证转染效率后,用划痕实验检测细胞迁移能力,应用Transwell小室检测侵袭迁移能力。Western blot检测过表达AGK后,SGC-7901细胞系中基质金属蛋白酶(MMP2/9)表达含量的变化。结果过表达AGK能够促进人胃癌细胞SGC-7901的迁移和侵袭能力。Western blot结果显示AGK的过表达能够上调胃癌细胞SGC-7901中的MMP2/9的蛋白表达水平。结论AGK上调胃癌细胞SGC-7901中MMP2/9的表达水平,促进胃癌细胞SGC-7901迁移和侵袭。Objective To explore the effects and mechanisms of lipid kinase ACK on the migration and invasion of human gastric cancer cell line SGC-7901.Methods Human gastric cancer SGC-7901 cells were cultured,and SGC-7901 cells were transiently transfected with AGK plasmid using HiGene reagent.After the transfection efficiency was verified,the healing rate of the scratch test measured the cell migration capacity.Transwell cells were used to detect migration and invasion capabilities.Western blot technology was used to detect changes in the expression levels of matrix metalloproteinase 2/9(MMP2/9)in overexpressed AGK,SCC-7901 cell line.Results The overexpression of AGK can promote the migration and invasion of human gastric cancer cells SGC-7901.Western blot results showed that overexpression of AGK can up-regulate the protein expression levels of MMP2/9 in gastric cancer cell SGC-7901.Conclusion In gastric cancer cell SGC-7901,AGK can up-regulate the expression level of MMP2/9,and promote the migration and invasion of gastric cancer cells.
