摘 要:目的探讨G蛋白信号调节剂14(RGS14)基因对缺氧缺糖再给氧诱导的大鼠海马神经元凋亡的影响及机制。方法构建RGS14-siRNA表达载体,转染入大鼠海马神经元内,实验分为对照组(Control)、缺氧缺糖再给氧模型组(Model)、RGS14基因沉默组(RGS14-siRNA)与转染阴性对照组(NC-RGS14)。RT-PCR检测RGS14基因表达,MTT检测细胞活力,TUNEL染色检测凋亡率,Western blot检测RGS14、p-TAK1、Caspase-3蛋白相对表达量。结果与Control组相比,Model组和NC-RGS14组RGS14基因及蛋白表达明显增加,细胞活力明显下降,p-TAK1、Caspase-3蛋白表达及细胞凋亡率明显增加(P<0.05);与Model组相比,RGS14-siRNA组RGS14基因及蛋白表达明显降低,细胞活力明显增加,p-TAK1、Caspase-3蛋白表达及细胞凋亡率明显降低(P<0.05)。结论沉默缺氧缺糖再给氧诱导的大鼠海马神经元RGS14基因可抑制细胞凋亡,其机制可能与抑制TAK1通路有关。Objective To investigate the effect and mechanism of regulator of G protein signaling 14(RGS14)gene on apoptosis of hippocampal neuron induced by hypoxia,glucose and reoxygenation in rats.Methods The eukaryotic expression vector of RGS14-siRNA was constructed and transfected into rat hippocampal neurons.The experiment was divided into four groups:control group(Control),hypoxia glucose deprivation reoxygenation model group(Model),RGS14 gene silencing group(RGS14-siRNA)and transfection negative control group(NC-RGS14).RT-PCR was used to detect the expression of RGS14 gene,MTT was used to detect cell viability,TUNEL staining was used to detect the apoptosis rate,and Western blot was used to detect the relative expression of RCS14,TAK1 and Caspase-3 proteins.Results Compared with control group,RGS14 gene and protein expression were significantly increased,cell viability was significantly decreased,TAK1 and Caspase-3 protein expression and apoptosis rate were significantly increased in Model group and NC-RGS14 group(P<0.05).Compared with Model group,the expression of RGS14 gene and protein in RGS14-siRNA group were decreased significantly,cell viability was increased significantly,and the expression of TAK1 and Caspase-3 protein and apoptosis rate were decreased significantly(P<0.05).Conclusion Silencing RGS14 gene induced by hypoxia,glucose and reoxygenation in rat hippocampal neurons can inhibit cell apoptosis,and its mechanism may be related to the inhibition of TAKl pathway.
