摘　　要：目的探究lncRNA XIST对甲状腺癌细胞增殖、侵袭和迁移能力的影响及可能机制。方法使用人类LncRNA Microarray V4.0芯片来分析甲状腺癌组织和旁侧正常组织LncRNA的差异性;观察lncRNAXIST与甲状腺癌患者临床资料的相关性;观察沉默lncRNAXIST对甲状腺癌细胞TPC-1和FTC133增殖、迁移和侵袭的影响;生物信息学预测lncRNAXIST的靶基因为miR-34a,荧光素酶验证其靶向结合位点,检测沉默lncRNAXIST对miR-34a表达的影响。结果lncRNA XIST在甲状腺癌中高表达,且与癌组织的分化程度、TNM分期及有无淋巴结转移呈正相关;lncRNA XIST靶向结合miR-34a;沉默lncRNA XIST可以抑制TPC-1和FTC133细胞的增殖、迁移和侵袭,并增强miR-34a的表达。结论lncRNA XIST靶向结合miR-34a并促进甲状腺癌细胞增殖、侵袭与迁移。Objective To explore the effects and possible mechanisms of IncRNA XIST on the proliferation,invasion and migration of thyroid cancer cells.Methods Human LncRNA Microarray V4.O chip was used to analyze LncRNA differences between thyroid cancer tissue and peripheral normal tissue.The correlation between lncRNAXIST and clinical data of patients with thyroid cancer was observed.The effects of silenced IncRNAXIST on the proliferation,migration and invasion of thyroid cancer cells TPC-1 and FTC133 was observed.Bioinformatics predicted that the target gene of lncRNAXIST was miR-34a,and luciferase verified its targeted binding site to detect the effect of silencing IncRNAXIST on the expression of miR-34a.Results IncRNA XIST was highly expressed in thyroid cancer,and was positively correlated with the differentiation degree of cancer tissue,TNM stage and lymph node metastasis.lncRNA XIST targeted miR-34a.Silencing IncRNA XIST could inhibit the proliferation,migration and invasion of TPC-1 and FTC133 cells,and enhance the expression of miR-34a.Conclusion LncRNA XIST targets miR-34a and promotes thyroid cancer cell proliferation,invasion and migration.
