摘 要:目的探讨缺氧预处理星形胶质细胞外泌体对脊髓神经元氧糖剥夺损伤的保护作用。方法神经元分为对照组、氧糖剥夺组、星形胶质细胞外泌体组、缺氧预处理星形胶质细胞外泌体组,除对照组外各组均进行神经元的氧糖剥夺,分别给予等体积PBS、正常细胞外泌体、缺氧预处理细胞外泌体,收集细胞,Western blot方法检测Bax、Caspase 3、Bcl-2表达,流式细胞术检测神经元凋亡率。结果神经元氧糖剥夺后,细胞凋亡率明显上升。与氧糖剥夺组相比,进行外泌体处理能明显降低神经元凋亡率及Bax、Caspase 3的表达、增加Bcl-2表达,其中缺氧预处理星形胶质细胞外泌体组保护作用更强。结论星形胶质细胞外泌体在脊髓神经元氧糖剥夺损伤中具有保护作用,缺氧预处理能增强这种保护作用。Objective To investigate the protective effect of hypoxia preconditioned astrocyte exosomes on spinal neurons in SD rats with oxygen glucose deprivation injury.Methods The neurons were divided into control group,oxygen glucose deprivation group,astrocyte exosomes group and hypoxia precondition astrocyte exosomes group.In addition to the control group,all groups were subjected to oxygen glucose deprivation of neurons,and were given equal volume PBS,normal cellular exosomes,and hypoxic pretreated cellular exosomes,respectively.The expressions of Bax,caspase-3 and Bcl-2 in cells were detected by Western blot,and apoptosis rate of cells was measured by flow cytometry.Results After oxygen and sugar deprivation,the apoptosis rate of neurons was increased significantly.Compared with the oxygen-glucose deprivation group,exosome treatment can significantly reduce apoptosis rate of the neurons,the expressions of Bax and Caspase 3 were decreased,the expression of Bcl-2 was increased,and the hypoxia pretreatment astrocyte exosome group had a stronger protective effect.Conclusion The hypoxia precondition enhanced the protective effect of astrocyte exosomes on the spinal neurons of oxidative glucose deprivation injury.
