摘  要:目的探讨肌动蛋白结合蛋白anillin(ANLN)作为肺腺癌新的治疗靶点的可能性,实验验证ANLN对肺腺癌细胞恶性表型的影响。方法采用免疫组织化学染色检测ANLN的表达,分析其对肺腺癌患者生存期的影响。在肺腺癌细胞株A549中敲低了ANLN表达,采用Western blot法检测敲减效率,划痕实验和Transwell实验分别检测ANLN对A549细胞迁移和侵袭的影响,集落形成实验检测ANLN对A549细胞增殖的影响。结果ANLN阳性表达的肺腺癌患者的累计生存率低于ANLN阴性表达的肺腺癌患者(P<0.01)。敲低ANLN表达可显著抑制A549细胞的侵袭、迁移和增殖(P<0.01)。结论在肺腺癌患者中,ANLN高表达可能预示患者生存期较差。ANLN有望成为治疗肺腺癌的潜在靶点。Objective To investigate the possibility of agonistic protein binding protein anillin(ANLN)as a new target gene for treating lung adenocarcinoma and experimentally validate its effect on the malignant phenotype of lung adenocarcinoma cells.Methods The expression of ANLN was detected with immunohistochemical experiments.The effect of ANLN expression on the overall survival of lung adenocarcinoma patients was analyzed.The ANLN expression in A549 cells was knocked down and its efficiency of knocking-down was detected by Western blot.Then the migratory and invasion of the A549 cells were detected by wound healing assay and Transwell assay,respectively.The colony formation assay was used to explore the effect of ANLN on the proliferation of A549 cells.Results The accumulative survival rate of the patients with positive expression of ANLN was lower than that of the patients with negative expression of ANLN(P<0.01).Knockdown of ANLN expression can significantly inhibit the invasion,migration and proliferation of lung adenocarcinoma cells(P<0.01).Conclusion In the patients with lung adenocarcinoma,high expression of ANLN often predicts a poor long-term survival.ANLN is hopeful to become a potential target for the treatment of the patients with lung adenocarcinoma.
