无患子皂苷的提取工艺及生物活性研究

摘　　要：为更好地利用无患子,研究无患子皂苷的提取工艺及生物活性。通过无患子果皮粉碎、溶液抽滤、旋转蒸发等过程获取无患子皂苷粗提物;针对粗提物,利用正丁醇等试剂,通过溶解、分层萃取与旋转蒸发等过程获取无患子皂苷精产物;选取齐墩果酸为标准品绘制标准曲线,计算皂苷含量、得率与纯度。利用光电比色计确定皂苷去污力,通过试管二倍稀释法检测抑菌活性,基于羟自由基作用测定方法测试抗氧化性。实验结果显示:原料粉碎处理且粒度为20~60目条件下,无患子皂苷纯度与产率更高;无水乙醇溶液浓度为70%时,皂苷得率达到上限;萃取比例为112时皂苷含量最高;发酵后皂苷液去污性能与抑菌性能高于皂苷浓缩液;无患子皂苷抗氧化活性较好。The extraction process and biological activity of soapberry saponin were studied in order to make better use of soapberry.The crude extract of soapberry saponin was obtained by the process of soapberry peel powder,solution suction filtration,rotary evaporation and other processes.N-butanol and other reagents were used to obtain soapberry saponin essence products through dissolution,layered extraction and rotational evaporation for crude extracts.Oleanolic acid was selected as the standard to draw a standard curve,and the saponin content,yield and purity were calculated.The saponin detergency detergency was determined by photoelectric colorimetry,the antibacterial activity was detected by the twofold dilution method of the test tube,and the antioxidant activity was tested based on the determination method of hydroxyl radical action.The experimental results showed that the purity and yield of soapberry saponin are higher under the condition of raw material powder treatment and particle size of 20~60 mesh.When the concentration of absolute ethanol solution is 70%,the yield of saponin reaches the upper limit.When the extraction ratio is 112,the saponin content is the highest.The decontamination and antibacterial properties of saponin solution are higher than those of saponin concentrate after fermentation.The antioxidant activity of soapberry saponins is good.
