摘  要:目的 使用中心体相关基因构建肝细胞癌预后模型,探讨其在预后和治疗中的意义。方法 从TCGA数据库和ICGC数据库中下载肝细胞癌数据集。使用单因素Cox分析鉴定出中心体相关预后基因,之后采用LASSO Cox回归构建预后模型。对高风险组和低风险组进行功能富集分析、免疫浸润分析、化疗和免疫治疗的反应预测。结果 通过单因素Cox分析鉴定出46个预后基因,之后使用LASSO Cox回归构建一个8基因预后模型。训练组和验证组的Kaplan-Meier分析均显示高风险组的预后更差。训练组1年、2年、3年的曲线下面积分别为0.789,0.696,0.692,验证组1年、2年、3年的曲线下面积分别为0.689,0.750,0.735。功能富集分析显示,高风险组和低风险组之间的差异基因主要与癌症和中心体相关。高风险组拥有更高的免疫浸润水平,并且对免疫检查点抑制剂和化疗药物的治疗更敏感。结论 使用中心体相关基因构建了预后模型,能够预测肝细胞癌患者的预后以及对化疗和免疫治疗的敏感性。Objective To construct a prognostic model for hepatocellular carcinoma by centrosome-associated genes and explore its significance in prognosis and treatment.Methods Download the hepatocellular carcinoma dataset from the TCGA and ICGC databases.Centrosome-associated prognostic genes were identified using univariate Cox analysis,after which LASSO Cox regression was used to construct prognostic models.Functional enrichment analysis,immune infiltration analysis,and response prediction to chemotherapy and immunotherapy were performed for the high-risk and low-risk groups.Results Forty-six prognostic genes were identified by univariate Cox analysis,after which an 8-gene prognostic model was constructed using LASSO Cox regression.Kaplan-Meier analysis for both the training and validation groups showed a worse prognosis for the high-risk group.The area under the curve for the training group were 0.789,0.696,and 0.692 for 1,2,and 3 years,respectively.For the validation group were 0.689,0.750,and 0.735 for 1,2,and 3 years,respectively.KEGG and GO analyses showed that differentially expressed genes between the high-risk and low-risk groups were mainly associated with cancer and centrosomes.The high-risk group had higher levels of immune infiltration and were more sensitive to treatment with immune checkpoint inhibitors and chemotherapeutic agents.Conclusion A prognostic model was constructed using centrosome-associated genes that were able to predict the prognosis of patients with hepatocellular carcinoma as well as their sensitivity to chemotherapy and immunotherapy.
