摘 要:基于甘草酸二铵与三苯甲烷类阳离子染料乙基紫发生离子缔合反应,首次提出离子缔合褪色反应分光光度法间接测定甜味剂甘草酸含量的新方法。在pH 8.6的Clark-Lubs缓冲溶液中,甘草酸二铵与乙基紫发生离子缔合反应,生成的离子缔合物使溶液显著褪色,褪色的最大吸收波长在594 nm处,据此建立了间接测定甘草酸的分析方法。结果显示,甘草酸二铵的浓度在3.00~12.00 mg/L之间与吸光度差值ΔA呈良好的线性关系,相关系数为0.9995,检出限为0.46 mg/L,表观摩尔吸光系数为3.20×10^(4)L/(mol·cm)。方法成功用于市售凉茶样品的含量测定,加标平均回收率为97.11%~101.0%,测定值的相对标准偏差RSD为0.59%~3.23%(n=5)。该法灵敏度高、专属性强、省时简便,可用于实际样品分析。Based on the ion association reaction between diammonium glycyrrhizinate and triphenylmethane cationic dye ethyl violet,a new method for the indirect determination of sweetener glycyrrhizic acid content by ionic association fading reaction spectrophotometry is proposed for the first time.In a Clark-Lubs buffer solution with pH 8.6,there is an ionic association reaction between diammonium glycyrrhizinate and ethyl violet,and the generated ion association compounds significantly fade the solution.The maximum absorption wavelength for fading is at 594 nm,and an indirect determination method for glycyrrhetinic acid has been established based on this.The results show that the concentration of diammonium glycyrrhizinate has a good linear relationship with the absorbance differenceΔA between 3.00 mg/L and 12.00 mg/L with a correlation coefficient of 0.9995,the detection limit of 0.46 mg/L and the apparent molar absorbance coefficient of 3.20×10^(4)L/(mol·cm).The method is successfully used for the determination of the content of commercially available herbal tea samples with the average spiked recovery rate of 97.11%~101.0%and the relative standard deviation(RSD)of the measured value of 0.59%~3.23%(n=5).This method has high sensitivity,strong specificity,is time-saving and convenient,and can be used for actual sample analysis.
