摘　　要：目的探索潜在转化生长因子β结合蛋白1(latent transforming growth factor beta binding protein 1,LTBP1)在胃癌发生发展及免疫微环境中的生物学功能。方法在TCGA数据库中分析LTBP1在泛癌中的表达,然后通过胃癌组织和癌旁组织进行验证。通过回归比例分析法分析LTBP1表达与临床病理变量之间的相关性。Cox回归分析和Kaplan-Meier图用于评估LTBP1在胃癌中的预后价值。通过TIMER分析LTBP1的表达水平与胃癌中免疫细胞浸润之间的相关性。免疫组织化学染色检测LTBP1蛋白在胃癌组织及癌旁组织中的表达水平。结果与正常胃组织相比,胃癌组织中LTBP1表达显著上调。其表达与病理TNM分期显著相关。LTBP1高表达的胃癌患者的总体生存率(overall survival,OS)缩短。免疫组化结果显示,与癌旁组织相比,LTBP1在胃癌组织中显著高表达。TIMER检测发现LTBP1的表达与3种免疫细胞浸润呈正相关。结论LTBP1可能是胃癌预后的一个潜在生物标志物,并影响癌症的肿瘤免疫微环境。Objective To explore the biological functions of latent transforming growth factor beta binding protein 1(LTBP1)in the occurrence,development,and immune microenvironment of gastric cancer.Methods The expression level of LTBP1 in gastric cancer was analyzed by using TCGA databases,the results were then validated in the gastric cancer tissues and the para-cancerous tissues.The association between LTBP1 expression level and the clinicopathological variables were analyzed by regression proportional analysis method.Cox regression analysis and Kaplan-Meier plots were used to assess the prognostic value of LTBP1 in gastric cancer.The correlation between the expression level of LTBP1 and the immune cells infiltration was analyzed by TIMER.Immunohistochemical staining was used to detect the expression level of LTBP1 protein in gastric cancer tissues and adjacent tissues.Results LTBP1 was significantly up-regulated in the gastric cancer tissues when compared with that in the normal samples.Its expression level was significantly correlated with the pathologic TNM stages.Higher LTBP1 expression indicated poor overall survival(OS)in patients with gastric cancer.We identified that LTBP1 expression had a positive correlation with 3 kinds of immune cells infiltration by TIMER.Immunohistochemical results showed that LTBP1 was significantly overexpressed in gastric cancer tissues.Conclusion LTBP1 can be a potential prognostic biomarker for gastric cancer patients and influences tumor immune microenvironment in gastric cancer.
