摘 要:目的探讨肌球蛋白重链11(MYH11)对人宫颈癌Hela细胞凋亡、迁移和侵袭作用的影响。方法应用实时荧光定量聚合酶联反应法(qRT-PCR)检测宫颈癌Hela细胞与正常宫颈上皮细胞Ect1/E6E7中MYH11的信使RNA(mRNA)表达水平,通过流式细胞术、噻唑蓝(MTT)实验及Transwell实验测定过表达MYH11对宫颈癌细胞凋亡、增殖、迁移和侵袭的影响,两组间比较采用t检验。结果qRT-PCR法检测mRNA表达,结果显示,宫颈癌Hela细胞中MYH11表达(0.52±0.04)明显低于正常宫颈上皮细胞(1.00±0.03),差异有统计学意义(t=10.56,P<0.01)。细胞凋亡实验结果显示,过表达MYH11组Hela细胞抗凋亡能力较ctrl组以及NC组表达显著降低。ctrl组、NC组、过表达MYH11组为(20.27±1.48)%、(19.83±1.85)%、(34.21±2.75)%,差异有统计学意义(F=45.69,P<0.01)。MTT增殖实验结果显示,Hela细胞中ctrl组、NC组以及过表达MYH11组24、48、72 h细胞增殖率分别为(54.28±2.89)%、(53.47±4.01)%、(40.25±2.87)%;(72.58±4.57)%、(72.78±4.06)%、(61.45±3.88)%;(87.56±5.69)%、(86.96±4.80)%、(72.52±4.83)%。过表达MYH11组增殖能力较ctrl组以及NC组表达显著降低,差异有统计学意义(F=17.09、7.22、8.29,P<0.05)。Transwell细胞迁移实验结果显示,过表达MYH11组细胞迁移能力[(52.48±5.01)个]明显低于ctrl组[(89.38±7.03)个]、NC组[(88.75±6.02)个],差异有统计学意义(F=36.26,P<0.01)。Transwell细胞侵袭实验结果显示,过表达MYH11组细胞侵袭能力[(18.56±2.41)个]明显低于ctrl组[(38.41±3.05)个]、NC组[(36.74±2.89)个],差异有统计学意义(F=46.50,P<0.01)。结论MYH11基因抑制宫颈癌Hela细胞的迁移和侵袭能力,促进细胞凋亡。Objective To investigate the effect of myosin heavy chain 11(MYH11)on apoptosis,migration and invasion of human cervical cancer HELA cells.Methods Real-time fluorescent quantitative polymerase chain reaction(RT-qPCR)was used to detect the mRNA expression level of MYH11.The effects of overexpression of MYH11 on apoptosis,proliferation,migration and invasion of cervical cancer cells were determined by flow cytometry,methyl thiazolyl tetrazolium(MTT)assay and Transwell assay.T-test was used for comparison between the two groups.Results The expression of mRNA was detected by RT-qPCR.The results showed that the expression of MYH11 in Hela cells of cervical cancer(0.52±0.04)was significantly lower than that in normal cervical epithelial cells(1.00±0.03),and the difference was statistically significant(t=10.56,P<0.01).The results of apoptosis experiment showed that the overexpression of MYH11 group had significantly higher apoptotic ability than Ctrl group and NC group[(20.27±1.48)%,(19.83±1.85)%,(34.21±2.75)%,F=45.69,P<0.01].MTT proliferation test results showed that the cell proliferation rates of Ctrl group,NC group and overexpression MYH11 group were(54.28±2.89)%,(53.47±4.01)%,(40.25±2.87)%;(72.58±4.57)%,(72.78±4.06)%,(61.45±3.88)%;(87.56±5.69)%,(86.96±4.80)%,(72.52±4.83)%,respectively,in 24,48 and 72 h of Hela cells.compared with Ctrl group and NC group,the proliferation ability of overexpression MYH11 group decreased significantly(F=17.09,7.22,8.29,P<0.05).Transwell cell migration experiment showed that the cell migration ability of the overexpression MYH11 group[(52.48±5.01)cells]was significantly lower than that of the Ctrl group[(89.38±7.03)cells]and NC group[(88.75±6.02)cells],and the difference was statistically significant(F=36.26,P<0.01).Transwell cell invasion experiment showed that the invasion ability of cells in the overexpression MYH11 group[(18.56±2.41)cells]was significantly lower than that in the Ctrl group[(38.41±3.05)cells]and NC group[(36.74±2.89)cells,F=46.50,P<0.01].Conclusi
