摘　　要：目的探讨子宫内膜癌组织中miR-141-3p、miR-149-3p表达及与增殖基因、临床病理特征和预后的关系。方法选取2017年2月至2019年2月该院诊治的98例子宫内膜癌患者作为研究对象。采用实时荧光定量PCR检测子宫内膜癌患者癌组织中miR-141-3p、miR-149-3p及增殖基因[增殖细胞核抗原(PCNA)、细胞周期素D1(cyclin D1)、细胞周期蛋白依赖激酶4(CDK4)]mRNA相对表达水平。Pearson相关分析子宫内膜癌患者癌组织中miR-141-3p、miR-149-3p表达与PCNA、cyclin D1、CDK4 mRNA表达的相关性。不同临床病理特征的子宫内膜癌患者癌组织miR-141-3p、miR-149-3p表达进行比较。Kaplan-Meier曲线分析子宫内膜癌患者癌组织miR-141-3p、miR-149-3p表达对其预后的影响。单因素及多因素Cox回归分析影响子宫内膜癌患者预后的因素。结果子宫内膜癌患者癌组织中miR-141-3p、miR-149-3p、PCNA mRNA、cyclinD1 mRNA、CDK4 mRNA相对表达水平高于癌旁组织,差异有统计学意义(P<0.05)。子宫内膜癌患者癌组织中miR-141-3p、miR-149-3p表达与PCNA mRNA、cyclinD1 mRNA、CDK4 mRNA表达呈正相关(均P<0.05)。国际妇产科联盟(FIGO)分期Ⅲ期、有淋巴结转移的子宫内膜癌患者癌组织miR-141-3p、miR-149-3p相对表达水平分别高于FIGO分期Ⅰ~Ⅱ期、无淋巴结转移的子宫内膜癌患者,差异有统计学意义(P<0.05)。miR-141-3p高表达组3年总体生存率显著低于miR-141-3p低表达组,差异有统计学意义(Log-rankχ^(2)=7.043,P=0.008)。miR-149-3p高表达组患者3年总体生存率显著低于miR-149-3p低表达组,差异有统计学意义(Log-rankχ^(2)=7.094,P=0.007)。FIGO分期Ⅲ期、有淋巴结转移、miR-141-3p升高、miR-149-3p升高是影响子宫内膜癌患者预后的独立危险因素(P<0.05)。结论检测子宫内膜癌患者癌组织中miR-141-3p、miR-149-3p表达有助于评估其患者生存预后。Objective To investigate the relationship between the expression of miR-141-3p,miR-149-3p and proliferative genes,clinicopathological features and prognosis in endometrial carcinoma tissues.Methods From February 2017 to February 2019,98 patients with endometrial cancer were selected as the study objects.Real-time fluorescence quantitative PCR was used to detect the relative mRNA expression levels of miR-141-3p,miR-149-3p and proliferating genes[proliferating cell nuclear antigen(PCNA),cyclin D1,cyclin dependent kinase 4(CDK4)]in cancer tissues of patients with endometrial cancer.The correlation between the expressions of miR-141-3p,miR-149-3p and PCNA,cyclin D1 and CDK4 mRNA in cancer tissues of patients with endometrial cancer was analyzed by Pearson correlation analysis.The expression of miR-141-3p and miR-149-3p in cancer tissues of endometrial carcinoma patients with different clinicopathological characteristics was compared.Kaplan-Meier curve was used to analyze the influence of miR-141-3p and miR-149-3p expression on prognosis of patients with endometrial carcinoma.Univariate and multivariate Cox regression analysis of prognostic factors in patients with endometrial cancer.Results The relative expression levels of miR-141-3p,miR-149-3p,PCNA mRNA,cyclinD1 mRNA and CDK4 mRNA in cancer tissues of patients with endometrial carcinoma were higher than those in adjacent tissues,with statistical significance(P<0.05).The expressions of miR-141-3p and miR-149-3p were positively correlated with the expressions of PCNA mRNA,cyclinD1 mRNA and CDK4 mRNA in cancer tissues of patients with endometrial carcinoma(all P<0.05).The relative expression levels of miR-141-3p and miR-149-3p in cancer tissues of endometrial cancer patients with stageⅢand lymph node metastasis according to International Federation of Gynecology and Obstetrics(FIGO)were higher than those of endometrial cancer patients with stageⅠ-Ⅱand no lymph node metastasis,respectively,and the difference was statistically significant(P<0.05).The 3-year overal
