circFOXK2靶向miR-409-3p调控肺癌细胞的增殖、迁移及侵袭

摘　　要：目的 探讨circFOXK2对肺癌细胞增殖、迁移和侵袭的影响及可能机制。方法 收集45例肺癌患者的癌组织和癌旁组织,采用qRT-PCR法检测组织中circFOXK2和miR-409-3p表达。体外培养肺癌细胞A549和H1299,双荧光素酶报告基因实验验证circFOXK2和miR-409-3p的调控关系。转染circFOXK2小干扰RNA、miR-409-3p模拟物、或共转染circFOXK2小干扰RNA与miR-409-3p抑制剂至A549和H1299细胞。细胞增殖、迁移和侵袭用CCK-8法、克隆形成实验和Transwell分析。蛋白质印迹法检测细胞中E-cadherin和N-cadherin蛋白表达。结果 肺癌组织中circFOXK2水平明显上升(P<0.05),而miR-409-3p水平明显下降(P<0.05)。circFOXK2在A549和H1299细胞中靶向结合并负调控miR-409-3p。敲减circFOXK2或过表达miR-409-3p后,肺癌细胞增殖、迁移和侵袭能力显著下降(均P<0.05),N-cadherin水平降低(P<0.05),而E-cadherin水平上升(P<0.05)。circFOXK2敲低对A549和H1299细胞增殖、迁移和侵袭的抑制作用可以被miR-409-3p抑制剂逆转。结论 circFOXK2在肺癌中表达升高,其通过靶向miR-409-3p促进肺癌细胞增殖、迁移和侵袭。Objective To investigate the effect of circFOXK2on the proliferation,migration and invasion of lung cancer cells and its possible mechanism.Methods The cancer tissues and adjacent tissues of 45patients with lung cancer were collected,and the expression of circFOXK2and miR-409-3p in the tissues were detected by qRT-PCR.Lung cancer cell A549and H1299was cultured in vitro,and the dual-luciferase reporter assay was used to verify the regulatory relationship between circ-FOXK2and miR-409-3p.The CCK-8,clone formation and Transwell assays were used to detect cell proliferation,migration and invasion.Western blot was used to detect the protein expression of E-cadherin and N-cadherin in cells.Results The expression of circFOXK2was significantly higher(P<0.05),but the expression of miR-409-3p was significantly lower(P<0.05)in lung cancer tissues.circFOXK2could target miR-409-3p and negatively regulate its expression in lung cancer cells.The proliferation,migration and invasion were reduced in circFOXK2(all P<0.05),N-cadherin level was decreased(P<0.05),but E-cadherin level was increased(P<0.05)in circFOXK2knockdown or miR-409-3p overexpressed A549and H1299cells.The inhibitory effects of circFOXK2knockdown on the proliferation,migration and invasion of lung cancer cells could be reversed by miR-409-3p inhibitor.Conclusion The circFOXK2was upregulated in lung cancer,which promoted the proliferation,migration and invasion of lung cancer cells by targeting miR-409-3p.
