摘 要:目的探讨血清外泌体microRNA-451a(miR-451a)是否可以作为结直肠癌(CRC)早期诊断及治疗的生物标志物。方法通过表达谱芯片筛选出CRC患者术前与术后血清外泌体中差异表达的miRNAs;利用肿瘤与癌症基因图谱(TCGA)数据库分析miR-451a在CRC组织中的表达并分析其临床意义;利用受试者工作特征(ROC)曲线下面积(AUC)分析miR-451a对CRC的诊断效能。生物信息学软件预测miR-451a的靶基因并进行基因本体(GO)和京都基因与基因组百科全书(KEGG)通路富集分析。结果miR-451a高表达与直肠癌患者远处转移呈负相关(r=-0.179,P=0.032)。miR-451a表达量对CRC组织具有较好的诊断效能(P<0.01)。生信分析结果显示miR-451a的靶基因与CRC发生发展的多种重要通路密切相关。结论外泌体miR-451a可作为CRC早期诊断的潜在生物标志物,辅助临床进行CRC早期诊疗。Objective To investigate the potential role of exosomal microRNA-451a(miR-451a)derived from serum in early diagnosis and treatment of colorectal cancer(CRC).Methods MiRNA microarry was performed to detect the miRNAs differently expressed between preoperative and postoperative serum exosomes derived from CRC patients.TCGA databases was used to analyze the expression levels of miR-451a in CRC tissues samples,and the correlation between miR-451a expression level and clinicopathological indices was analyzed.The area under receiver operating characteristic(ROC)curve(AUC)was used to analyze the diagnostic efficacy of miR-451a in rectal cancer and colon cancer,respectively.The target genes of miR-451a with enriched pathways were analyzed by using bioinformatics software.Results The high expression level of miR-451a was negatively correlated with distant metastasis in rectal cancer patients(P=0.032,r=-0.179).The expression level of miR-451a had a good diagnostic capability in CRC tissues(P<0.01).Enrichment analyses revealed that the target genes of miR-451a were closely related to several important pathways in the occurrence and development of CRC.Conclusion Exosomes miR-451a might be a potential biomarker for the early diagnosis and treatment of CRC.
