摘 要:蚕丝精炼废水中的丝胶蛋白增加了污水治理的难度,而将其制备成抗菌材料是目前废丝胶蛋白资源利用的常用方法。在以前的研究中一般是使用纯化后的丝胶蛋白与银离子等掺杂进行抗菌研究,此类抗菌材料中丝胶蛋白纯化的成本很高,且与其他抗菌材料复合中丝胶蛋白的使用量不高,主要是银离子等起到抗菌作用,这很可能将大大限制废丝胶蛋白“变废为宝”的道路。本文为了探索丝胶蛋白粗品在纯化前的抗菌性能,选用精炼废液中浓缩回收的丝胶蛋白粉,研究其本身通过沉降溶解多次提纯后及高纯度低分子量的商售丝胶蛋白对大肠杆菌和金黄色葡萄球菌的抑菌情况。抑菌圈法和菌落法的结果都表明,同一分子量丝胶蛋白对大肠杆菌的抑菌效果优于金黄色葡萄球菌,且随着分子量的提高,抗菌能力增加。结果还显示,在一定纯度范围内,分子量是决定丝胶蛋白的主要因素,而不是纯度,因为提纯四次后的丝胶蛋白的抑菌性比未提纯前只有微小的提升。研究结果可为未经纯化的丝胶蛋白粗品的低成本抑菌应用提供思路。The sericin in the silk refining wastewater has increased the difficulty of wastewater treatment,and preparing it into antibacterial materiall is a common mothod to use waste sericin resources.In the previous studies,the purified sericin was generally mixed with silver ions for antibacterial research.The cost of sericin purification in this kind of antibacterial material is very high,and the use of sericin in combination with other antibacterial materials is not high,mainly because silver ions play an antibacterial role,which is likely to greatly limit the way of“turning waste sericin into treasure”.In order to explore the antibacterial performance of crude sericin before purification,sericin powder concentrated and recovered from refining waste liquid was selected to study the bacteriostasis of sericin on Escherichia coli and Staphylococcus aureus by itself,after multiple purification through sedimentation and dissolution,and commercial sericin with high purity and low molecular weight.The results of inhibition zone method and colony method showed that the antibacterial effect of sericin with the same molecular weight on Escherichia coli was better than that of Staphylococcus aureus,and the antibacterial ability increased with the increase of molecular weight.The results also showed that in a certain purity range,the molecular weight was the main factor determining the sericin,not the purity,because the bacteriostasis of sericin after four times of purification was only slightly improved than that before purification.The results can provide some ideas for the low-cost antibacterial application of crude sericin without purification.
