摘  要:目的基于网络药理学探讨瓜子金对脑缺血疾病治疗的作用机制。方法通过TCMSP数据库挖掘出瓜子金中所含有的化学有效成分及作用靶点并经Uniprot数据库标准化,运用Cytoscape3.8.0软件构建活性成分-关键靶点网络可视图。再利用OMIM、TTD、TCMSP、ETCM、DisGeNET和GeneCards数据库检索脑缺血相关基因,利用韦恩图筛选出瓜子金中活性成分治疗脑缺血的作用靶点,利用Cytoscape3.8.0软件构建药物-疾病-靶点网络可视图。利用R语言cluster Profiler包进行蛋白互作用PPI网络分析,以及GO功能和KEGG通路富集分析,并验证网络分析结果。结果从瓜子金所含59个皂苷成分和41个黄酮类成分中筛选得到4个活性化合物及147个潜在靶点,4个活性成分分别是常春藤皂苷元、槲皮素、山奈酚、1,3,7-三羟基-9H-氧杂蒽-9-酮。脑缺血中相关靶点有1671个,利用VENN图筛选出药物-疾病共同靶点105个。GO功能富集分析,包括生物学过程119条;KEGG通路富集分析显示163条通路,主要涉及血脂和动脉粥样硬化、糖尿病并发症的AGE-RAGE信号通路、肿瘤坏死因子信号通路、IL-17信号通路等。结论瓜子金对治疗脑缺血具有多成分、多靶点和多通路的作用特点,为瓜子金临床应用治疗脑缺血提供了思路和方法。Objective:To explore the action mechanism of Polygalae Japonica in the treatment of cerebral ischemia disease based on network pharmacology.Methods:The chemical active components and their target sites in Polygalae Japonica were extracted from TCMSP database and standardized by Uniprot database.The active component-key-target network was constructed by using Cytoscape3.8.0 software.And OMIM,TTD,TCMSP,ETCM,DisGeNET and GeneCards databases were used to search for cerebral ischemia-related genes and then the target of the active components in Polygalae Japonica for treating cerebral ischemia was selected using Venn diagram.The medicine-disease-target network was constructed using Cytoscape3.8.0 software.The R language cluster Profiler package was used for protein interaction PPI network analysis,GO function and KEGG pathway enrichment analysis and the results of network analysis were verified.Results:From 59 saponins and 41 flavonoids contained in Polygalae Japonica,4 active compounds and 147 potential targets were selected.The 4 active compounds were hederagenin,quercetin,kaempferol and 1,3,7-hydroxy-9h-oxanthrace-9-ketone,respectively.There were 1671 related targets in cerebral ischemia and 105 common medicine-disease targets were screened by VENN diagram.The GO functional enrichment analysis included 119 biological processes and the KEGG pathway enrichment analysis showed 163 pathways,mainly involved in lipid and atherosclerosis,diabetes complications AGE-RAGE signaling pathway,tumor necrosis factor signaling pathway,IL-17 signaling pathway and so on.Conclusion:Polygalae Japonica has the characteristics of multi-component,multi-target and multi-pathway action in the treatment of cerebral ischemia,which provides ideas and methods for the clinical application of Polygalae Japonica in the treatment of cerebral ischemia.
