摘 要:目的:研究纳米银腹腔注射对小鼠H22肝癌腹水瘤的疗效及安全性。方法:昆明小鼠40只经腹腔注射H22细胞建立肝癌腹水瘤模型,24 h后随机分为模型组(生理盐水),顺铂组及纳米银低、中、高剂量组,连续12天腹腔注射给药,记录小鼠一般状况、体重及腹围,并观察腹水生成及生存情况。另选择健康昆明小鼠20只随机分为对照组及纳米银高剂量组进行药物安全性实验,连续12天腹腔注射相应药物,记录小鼠体重及生存情况,全自动血细胞分析仪、全自动生化分析仪检测小鼠血常规及生化指标。结果:与模型组相比,纳米银组小鼠体重及腹围增加缓慢,生存期延长,纳米银高剂量组小鼠体重和腹围变化及生存期与顺铂组相比差异无统计学意义。安全性实验结果显示,与对照组相比,纳米银高剂量组小鼠体重、生存期、血常规和肝肾功能各项生化指标的差异均无统计学意义。结论:纳米银可抑制肝癌腹水生成并延长腹水瘤小鼠生存期,且无明显毒副作用,或可作为癌性腹水治疗的一种新方法。Objective:To investigate the effectiveness and safety of silver nanoparticles(Ag-NPs)treating H22 hepatoma ascites of mice via intraperitoneal injection.Methods:H22 cells were intraperitoneally injected in 40 Kunming mice for 12 consecutive days to set up a hepatoma ascites model.The Kunming mice were,thereby,assigned to the model group(normal saline),the cisplatin group,the low-Ag-NPs group,the medium-Ag-NPs and the high-Ag-NPs group.We observed the performance status,weight,abdomen circumference,ascites growth and survival of the mice.Another 20 healthy mice injected intraperitoneally with corresponding drugs for 12 consecutive days were further assigned to the control group and the high-Ag-NPs group for drug safety assessment.The weight and survival of the mice were observed.Hematology and biochemical indexes of the mice were detected by automatic blood cell analyzer and automatic biochemical analyzer.Results:Compared to the model group,the weight and abdomen circumference increased slowly and the survival prolonged in the Ag-NPs groups,and there were no significant differences between the high-Ag-NPs group and the cisplatin group in weight,abdomen circumference and survival of the mice.The results of the drug safety assessment showed that there were no significant differences between the blank control group and the high-Ag-NPs group in weight,survival,hematology and biochemical indexes of the mice.Conclusion:Ag-NPs can suppress the growth of ascites and improve the survival of ascites mice without evident side effects,which can be a new method to cure hepatoma ascites.
